摘  要:目的检测长链非编码RNA(lncRNA)H19基因在甲状腺癌中的表达并探讨其表达差异与临床病理学参数及细胞增殖能力的关系。方法收集2016年5月至2017年6月河北医科大学第四医院接受手术治疗的甲状腺癌组织及癌旁组织标本30例,应用实时荧光定量聚合酶链反应(RT-qPCR)法检测甲状腺癌及其癌旁组织中lncRNA H19的表达水平,分析其与患者临床病理学参数间的关系;采用噻唑蓝(MTT)法检测敲低lncRNA H19组和阴性对照组对甲状腺癌细胞增殖能力的影响。组间比较采用χ^(2)或t检验分析。结果 lncRNA H19在甲状腺癌组织中表达水平(6.72±0.12)明显高于癌旁组织(1.09±0.08,t=7.494,P<0.01);lncRNA H19的表达水平与甲状腺癌患者的组织学分级显著相关(χ^(2)=6.205,P<0.05);MTT实验结果表明,转染lncRNA H19小干扰RNA(siRNA)的FTC133细胞48 h的吸光度(A)值(0.87±0.09)显著低于对照组细胞(1.12±0.13),差异有统计学意义(t=16.250,P<0.01)。结论 lncRNA H19在甲状腺癌组织中表达升高,并且与患者的分化程度明显相关,敲低lncRNA H19的表达能够明显抑制甲状腺癌细胞的增殖能力,这表明lncRNA H19在甲状腺癌的发生发展中起重要作用。Objective To elucidate the expression of long non-coding RNA(lncRNA)H19 gene in thyroid cancer and explore the relationship between its expression difference and clinicopathological parameters and cell proliferation ability.Methods Totally,30 cases of thyroid cancer tissues and paracancerous tissues that received surgical treatment in the fourth hospital of Hebei Medical University from May 2016 to June 2017 were collected,the real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)was used to detect the expression level of lncRNA H19 in thyroid cancer and its adjacent tissues,and analyze the relationship between lncRNA H19 and patient clinicopathological parameters;Thiazolium blue methyl thiazolyl tetrazolium(MTT)method was used to detect the effect of knockdown lncRNA H19 group and negative control group on the proliferation of thyroid cancer cells.Differences between groups were analyzed byχ^(2) or t test.Results The expression level of lncRNA H19 in thyroid cancer tissues(6.72±0.12)was significantly higher than that in adjacent tissues(1.09±0.08,t=7.494,P<0.01),the expression level of lncRNA H19 was significantly correlated with the histological grade of patients with thyroid cancer(t=6.205,P<0.05).The results of the MTT experiment showed that the 48 h absorbance(A)value of FTC 133 cells transfected with lncRNA H19 small interfering RNA(siRNA,0.87±0.09)was significantly lower than the 48 h A value of control group(1.12±0.13),and the difference was statistically significant(t=16.250,P<0.01).Conclusion The expression of lncRNA H19 is elevated in thyroid cancer tissues and is related to the degree of differentiation of patients.Knockdown of the expression of lncRNA H19 can significantly inhibit the proliferation of thyroid cancer cells,which indicates that lncRNA H19 plays an important role in the occurrence and development of thyroid cancer.
