摘  要:目的检测口腔癌常用的化疗药物5-FU、PTX、CDDP联合特比萘酚(tPrhinafine,TB)对癌细胞增殖的影响并初步探讨作用机制。方法体外培养CAL27、HN4、HN6、HN30等4个口腔鱗癌细胞系,化疗药物在0~30 nmol/L范围设置6个浓度,TB设置2个浓度:0μmol/L和10μmol/LMTT法检测细胞增殖,免疫印迹方法检测SQLE蛋白表达水平。结果与5-FU单独作用比较,联用TB时HN30和HN4细胞的增殖显著降低(P<0.05)。与PTX单独使用比较,联用TB时CAL27细胞的IC50降低了49.3%;HN30细胞的1C50降低了43.7%。与CDDP单独使用比较,联用TB时HN6细胞的IC50降低了65.8%,细胞增殖降低18.2%~58.3%(P<0.05),SQLE蛋白表达明显上升。与CDDP联用时,20μmol/LTB的抑制效果明显高于10μmol/L。结论TB联合5-FU、CDDP化疗药物可以提高对口腔鱗癌细胞增殖的抑制作用,其作用机制可能与TB抑制SQLE表达水平有关。Objective To investigate the effects of 5-Fu, PTX and CDDP combined with terbinafine(TB) on the proliferation and role of oral squamous cell carcinoma cells. Methods The oral squamous cell carcinoma(OSCC) cell lines CAL27, HN4, HN6 and HN30 were cultured in vitro. The concentration of chemotherapy drugs was set at 6 levels within the range of 0-30 μmol/L, and the concentration of TB was set at 2 levels, 0 μmol/L and 10 μmol/L. Cell proliferation was determined by MTT method and western blot method was applied to detect squalene epoxidase(SQLE) protein expression level. Results Compared with the treatment of 5-FU alone, the viabilities of HN30 and HN4 cells were decreased when treated with 5-FU together with TB. In contrast to PTX alone, the IC50 of PTX combined with TB in CAL27 and HN30 were reduced by 49.3% and 43.7%, respectively.The IC50 of CDDP and TB in HN6 was reduced by 65.8% compared with CDDP alone, and the viability of HN6 cells decreased by 18.2%-58.3%(P<0.05). The inhibitory effect of 20 μmol/L TB was significantly stronger than that of 10 μmol/L TB. SQLE protein expression levels of HN6 cells were significantly increased when CDDP was combined with TB. Conclusions Combinations of 5-FU or CDDP with TB may enhance the cell proliferation inhibition of oral squamous cell carcinoma cells, and the mechanism may be related to the inhibition of SQLE activity by TB.
