摘  要:目的评价沉默S100A12基因对甲状腺乳头状癌细胞K1的增殖、侵袭与迁移能力的影响。方法用shRNA质粒转染K1细胞沉默S100A12基因,将细胞分为shRNA-NC组和shS100A12组,通过MTT实验评价细胞增殖生长趋势,平板克隆形成实验检测细胞克隆形成数量,Transwell实验检测细胞侵袭迁移能力,细胞划痕实验评价细胞的迁移能力,Western blot实验检测K1细胞E-cadherin,N-cadherin蛋白的表达情况。结果与shRNANC组比较,shS100A12组K1细胞生长趋势明显受到抑制(P<0.01),细胞克隆形成数量明显下降(P<0.01),通过通透膜的细胞数量明显减少(P<0.05),细胞的迁移速率明显减慢(P<0.01)。与shRNA-NC组比较,shS100A12组细胞E-cadherin的表达增高、N-cadherin的表达下降。结论沉默甲状腺乳头状癌K1细胞中S100A12可以降低细胞增殖、侵袭与迁移能力,S100A12可能通过诱导EMT增强甲状腺乳头状癌细胞的侵袭与迁移能力。Objective To evaluate the effect of S100A12 gene silencing on proliferation, invasion and migration of papillary thyroid carcinoma cell line K1. Methods After being transfected with shRNA plasmid to silence the S100A12 gene, K1 cells were divided into shRNA-NC group and shS100A12 group. The MTT experiment was used to evaluate the proliferation and growth trend of the cells. The clone formation test was used to compare the number of cell clone formation. The Transwell test was used to compare the invasion and migration ability. The cell scratch test was used to evaluate the migration ability of the cells. Western blot test was used to detect E-cadherin and N-cadherin proteins in K1 cells. Results Compared with shRNA-NC group, the growth trend of cells in the shS100A12 group was significantly inhibited(P<0.01), the quantity of clone-forming cells in the shS100A12 group within the same time was significantly reduced(P<0.01), the quantity of the cells passed through the permeable membrane was significantly reduced(P<0.05), the migration rate of cells was significantly slowed down(P<0.01). Compared with shRNA-NC group, the expression of E-cadherin in the shS100A12 group was increased, and the expression of N-cadherin was decreased. Conclusion Silencing S100A12 can reduce the proliferation, invasion and migration of K1 cells. S100A12 may enhance the invasion and migration ability of papillary thyroid cancer cells by inducing EMT.
