摘 要:目的探讨miR-375对肝癌细胞生长和迁移的调控方式。方法通过PCR检测miR-375和HMGB3mRNA表达,通过Western blot检测HMGB3蛋白表达,荧光素酶实验检测分子间结合。CCK8实验检测不同处理因素下的细胞活力并通过Transwell实验检测细胞迁移能力。结果 miR-375在肝癌细胞系中低表达,而HMGB3在肝癌细胞中高表达,miR-375和HMGB3 mRNA之间存在结合位点。miR-375可抑制HMGB3的表达,减缓肝癌细胞的生长和迁移。结论 miR-375能抑制HMGB3,从而减弱肝癌细胞的生长和迁移能力。Objective To explore the mechanism of miR-375 on cell proliferation and migration. Methods The expression of miR-375 and HMGB3 mRNA was detected by PCR, and the expression of HMGB3 protein was detected by Western blot. The molecular binding sites were analyzed by luciferase assays. CCK8 and Transwell assays detected the cell viability and migration under different treatment factors. Results miR-375 was low-expressed in HCC cell lines(P < 0.05), while HMGB3 was highly expressed in HCC cells, and there were binding sites between miR-375 and HMGB3 mRNA. miR-375 can inhibit HMGB3 expression and slow down the growth and migration of HCC cells(P<0.05). Conclusion miR-375 can inhibit HMGB3 to weaken the growth and migration of HCC cells.
