摘 要:目的研究维生素D3对胃癌细胞SGC-7901细胞迁移的作用及分子机制。方法将胃癌细胞SGC-7901分为对照组和维生素D3组,分别用DMSO和维生素D3处理,用Transwell迁移实验检测胃癌细胞迁移能力。通过实时荧光定量PCR(qRT-PCR)检测正常胃黏膜上皮细胞GES-1和AGS、BGC-823、SGC-7901胃癌细胞系中维生素D受体(VDR)的表达情况。将胃癌细胞SGC-7901分为对照组、维生素D3组、miR-145抑制剂组和联合处理组,分别用DMSO、维生素D3、miR-145抑制剂、miR-145抑制剂转染加维生素D3处理,用Transwell迁移实验检测胃癌细胞迁移能力的变化。结果与对照组相比,维生素D3组迁移的胃癌细胞减少(P<0.01)。与正常胃黏膜上皮细胞相比,胃癌细胞系中维生素D受体表达降低(P<0.001)。与对照组相比,miR-145抑制剂组迁移的细胞增加(P<0.001);与维生素D3组相比,联合处理组胃癌细胞迁移的细胞增加(P<0.01)。结论维生素D3能够抑制胃癌细胞SGC-7901的迁移,维生素D受体在胃癌细胞中低表达,且维生素D3可能通过miR-145抑制胃癌细胞的迁移。Objective To study the effect of vitamin D3 on the migration of gastric cancer cells and its molecular mechanism.Methods SGC-7901 cells were divided into control group and vitamin D3 group,and were treated with DMSO and vitamin D3,respectively.Transwell assay was used to detect the migration ability of cells.The expression of vitamin D receptor in normal gastric mucosa epithelial cells GES-1 and gastric cancer cell lines AGS,BGC-823,SGC-7901 was detected by qRT-PCR.SGC-7901 cells were divided into control group,vitamin D3 group,miR-145 inhibitor group and combined treatment group,and were respectively treated with DMSO,vitamin D3,miR-145 inhibitor,and miR-145 inhibitor transfection+vitamin D3.Transwell assay was used to detect the migration ability of gastric cancer cells.Results Compared with control group,the migration ability of gastric cancer cells was decreased in vitamin D3 group(P<0.01).The vitamin D receptor expression was lower in gastric cancer cell lines than that in normal gastric epithelial cells(P<0.001).Compared with control group,the migration ability of cells in miR-145 inhibitor group was increased(P<0.001).Compared with vitamin D3 group,the migration of gastric cancer cells was increased in combined treatment group(P<0.01).Conclusion Vitamin D3 can inhibit the migration of gastric cancer SGC-7901 cells.Vitamin D receptor is lowly expressed in gastric cancer cells,and vitamin D3 may inhibit the migration of gastric cancer cells through miR-145.
