摘 要:目的探究乳腺癌FE1.2细胞高表达miR-24对Nod/Scid免疫缺陷小鼠移植瘤生长及肺转移的影响。方法通过脂质体转染法将高表达miR-24的载体及空载体分别转染大鼠乳腺癌FE1.2细胞,分别设为FE1.2-miR-24 hi组及FE1.2-v组,自身高表达miR-24的大鼠乳腺癌FE1.3细胞设为FE1.3组,采用qPCR实验检测各组miR-24的表达。将转染后的细胞分别接种于雌性Nod/Scid免疫缺陷小鼠的第4对乳腺脂肪垫,构建免疫缺陷鼠移植瘤模型,分别设为FE1.2-miR-24 hi组及FE1.2-v组,经免疫缺陷鼠移植瘤实验检测各组移植瘤生长及肺转移情况。结果qPCR实验结果显示,与FE1.2-v组及FE1.3组比较,FE1.2-miR-24 hi组miR-24表达水平显著升高(均P<0.01)。免疫缺陷鼠移植瘤实验结果显示,与FE1.2-v组比较,FE1.2-miR-24 hi组移植瘤体积和质量显著减小,肺部转移明显减少(均P<0.01)。结论miR-24能显著抑制Nod/Scid免疫缺陷小鼠中乳腺癌FE1.2细胞移植瘤生长和肺转移。Objective To explore the effects of miR-24 overexpression on growth and lung metastasis of breast cancer cell FE1.2 xenograft in Nod/Scid immunodeficient mice.Methods Rat FE1.2 cells were transfected with miR-24 plasmids and negative vector by Lipofectamine transfection,and the transfected cells were divided into FE1.2-miR-24 hi group and FE1.2-v group,respectively.Rat breast cancer FE1.3 cells with miR-24 overexpression were selected as FE1.3 group.The expression of miR-24 in each group was detected by qPCR.In addition,the transfected FE1.2-miR-24 hi and FE1.2-v cells were inoculated into the fourth pair of mammary fat pads of Nod/Scid immunodeficient mice to construct xenograft models of FE1.2 cells,respectively,which were named as FE1.2-miR-24 hi group and FE1.2-v group.And then xenograft growth and lung metastasis in each group were detected by xenograft model experiments.Results The results of qPCR showed that compared with FE1.2-v group and FE1.3 group,miR-24 expression was significantly increased in FE1.2-miR-24 hi group(P<0.01).Compared with FE1.2-v group,growth and lung metastasis of breast can-cer cell FE1.2 xenograft in Nod/Scid immunodeficient mice were significantly reduced in FE1.2-miR-24 hi group(P<0.01).Conclusion The miR-24 could significantly inhibit growth and lung metastasis of breast cancer cell FE1.2 xenograft in Nod/Scid immunodeficient mice.
