摘  要:目的评价转染有胰岛素样生长因子-1(IGF-1)基因的骨髓基质细胞(MSCs)复合鸵鸟钙磷陶瓷骨对狗缺损的牙周组织再生的影响及作用。方法将转染有IGF-1基因的狗自体MSCs与鸵鸟钙磷陶瓷骨(OTBC)复合,扫描电镜观察复合情况。选用3只狗,30牙,制备相同的牙周组织缺损,然后随机分为3组:T-MSCs/OTBC组、MSCs/OTBC组和OTBC组。T-MSCs/OTBC组将IGF-1基因转染MSCs细胞复合OTBC支架置入牙周缺损区;MSCs/OTBC组将MSCs细胞复合OTBC支架置入牙周缺损区;OTBC组仅采用单纯OTBC支架。术后8周处死狗,并对术牙切片染色,进行组织学观察和测量。结果扫描电镜观察发现,转染有IGF-1基因的狗自体MSCs与OTBC复合后生长良好,24 h细胞即可见贴附、伸展,48 h可见细胞生长与增殖,分泌有大量细胞外基质。狗牙组织学结果可见T-MSCs/OTBC组牙周组织再生较完全,没有结合上皮长入,未发现根吸收和骨粘连;MSCs/OTBC组大部分牙周组织缺损被修复,但可见少量结缔组织长入;OTBC组仅有少量的牙周组织被修复,有纤维结缔组织和长上皮细胞长入。进一步组织学测量结果显示,T-MSCs/OTBC组新生牙槽骨、牙骨质以及牙周结缔组织附着量与OTBC组比较差异均有统计学意义(P<0.05);与MSCs/OTBC组相比较,T-MSCs/OTBC组有较多的牙槽骨新生,差异有统计学意义(P<0.05),但牙骨质以及牙周结缔组织附着量差异无统计学意义(P>0.05)。结论IGF-1基因转染骨髓基质细胞复合鸵鸟钙磷陶瓷骨能有效地促进狗牙周缺损组织再生。Objective To evaluate the effect of bone marrow stromal cells(MSCs)transfected with insulin-like growth factor-1(IGF-1)gene combined with ostrich true bone ceramics on periodontal tissue regeneration in dogs.Methods The dog autogenous MSCs transfected with IGF-1 gene were recombined with ostrich true bone ceramics(OTBC),and the recombination was observed by scanning electron microscopy.Thirty teeth in three dogs were induced to establish periodontal tissue defect models and then randomly divided into three groups:T-MSCs/OTBC group,MSCs/OTBC group and OTBC group.The periodontal tissue defect area was implanted with MSCs transfected with IGF-1 gene recombined with OTBC in T-MSCs/OTBC group,the periodontal tissue defect area was implanted with MSCs recombined with OTBC in MSCs/OTBC group,and the periodontal tissue defect area was implanted with OTBC in OTBC group.Eight weeks after the operation,the dogs were sacrificed,and the dental sections were stained for histological observation and measurement.Results Scanning electron microscope observation showed that the recombination between OTBC and MSCs transfected with IGF-1 gene grew well.Adhesion and extension of cells were observed at 24 h,cell growth and proliferation were observed at 48 h,and a lot of extracellular matrix was secreted.Histological changes of dog teeth showed that the periodontal tissue regeneration in T-MSCs/OTBC group was complete,no junctional epithelium grew into the periodontal tissue defects,and no root absorption and bone adhesion were observed.In MSCs/OTBC group,most of the periodontal tissue defects were repaired,but still a small amount of connective tissue was found.In OTBC group,only a small amount of periodontal tissue was repaired,with fibrous connective tissue and long epithelial cells growing into the periodontal tissue defects.Histological measurements showed that there was significant difference in the amount of new alveolar bone,cementum and periodontal connective tissue attachment between T-MSCs/OTBC group and OTBC group(P<0.05).
