摘 要:目的建立海带多酚的提取及纯化工艺。方法以乙醇浓度、提取时间、提取温度和料液比进行单因素试验,并在单因素试验基础上采用4因素3水平进行正交试验优化提取工艺,同时采用树脂进行纯化工艺优化。结果确定最佳提取工艺为:采用85%乙醇为溶剂,以料液比1∶65,在65℃下搅拌提取1 h。在提取海带粗多酚后采用大孔吸附树脂进行分离纯化研究,筛选出最佳的大孔吸附树脂SZ-3,优化最佳层析条件,获得高纯度海带多酚(80%)。结论本方法简便可行,可用于海带多酚的提取与纯化。Objective To establish the extraction and purification process of Laminaria japonica polyphenols.Methods The Laminaria japonica polyphenol extraction process was investigated by single factor test with ethanol concentration,extraction time,extraction temperature and feed-liquid ratio.The orthogonal experiment was carried out to determine the optimization extraction condition.Then resin was used to optimize the purification process.Results The optimal condition was ethanol concentration 85%,the material solvent ratio 1∶65,under the temperature of 65℃for 1 h.After extracting crude polyphenols from Laminaria japonica,macroporous adsorption resin was used to separate and purify Laminaria japonica polyphenols.The macroporous adsorption resin suitable for purification of Laminaria japonica polyphenols.The Results showed that SZ-3 had got the best purification effect,and the optimum chromatographic conditions were optimized to obtain high purity Laminaria japonica polyphenols(80%).Conclusion The method was simple and feasible,and can be used for the extraction and purification of Laminaria japonica polyphenols.
