摘 要:类胡萝卜素是一种多烯类化合物,是生活中常见的一种色素,是食品的着色剂和营养强化剂。以山西运城盐湖中的一株嗜盐古菌为研究对象,采用无水乙醇作为有机溶剂对其产生的类胡萝卜素进行提取。选取提取时间、提取温度、料液比和提取次数作为因素,采用正交试验对嗜盐古菌产生的类胡萝卜素的提取工艺条件进行优化。结果表明,山西运城盐湖嗜盐古菌产生的类胡萝卜素提取工艺的最佳条件为最佳提取有机溶剂为无水乙醇,提取料液比1∶8,提取时间30 min,提取温度40℃,最佳提取次数为2次,类胡萝卜素提取含量最高可达183.07μg/g,为从山西运城盐湖嗜盐古菌中提取类胡萝卜素及扩大类胡萝卜素生产提供重要的科学依据。Carotenoids are Polynesia compounds,which are a common pigment in life.It is often used as a food additive colorant and nutrition enhancer.The research object was halophytic archaea in Yuncheng salt lake,Shanxi Province.The optimum extraction process of carotenoid was studied using Anhydrous Ethanol.The effects of temperature,time,ratio of material to liquid and times of extraction on the yield of carotenoid were studied by single factor experiments.The ratio of material to liquid had the greatest effect on the yield of carotenoids.The optimum extraction conditions of different factors were determined by Orthogonal Experiment.The results showed that the optimum conditions for extraction of carotenoids from halophytic archaea were as follows:The sample was halophytic archaea from Yuncheng salt lake,Shanxi Province,the best organic solvent was Anhydrous Ethanol,the ratio of extracted material to liquid was 1∶8,the extraction time was 30 minutes,the extraction temperature was 40 degrees Celsius and the number of extraction was 2 times.The extract content of carotenoids was up to 183.07μg/g.To find out the optimum technology,which will provide an important scientific basis for extracting carotenoid from Halophytic archaea in salt lake,Shanxi and expanding the production of carotenoid.
