摘  要:低氧是结直肠癌肿瘤组织的重要特征和结直肠癌预后不良的重要因素。然而,低氧如何调控直肠癌细胞的基因表达机制仍待深入分析。通过转录组学和翻译组学联合分析揭示了低氧条件下基因表达特征。首先利用化合物CoCl 2处理SW620细胞成功模拟了低氧条件。然后利用mRNA-seq和Ribo-seq分析发现,许多基因的转录和翻译会受低氧条件影响,且多个肿瘤相关的信号通路中的基因翻译效率也受低氧调控。另外,利用real-time PCR和Western blot技术对部分低氧调控的基因表达进行了验证。从新的角度阐明低氧调控肿瘤相关基因表达特征,有助于弄清直结肠癌的发生机制。Hypoxia is an important feature of colorectal cancer and an important factor for poor prognosis of colorectal cancer.However,the mechanism of how hypoxia regulates gene expression in rectal cancer cells remains to be further analyzed.In this study,through combining transcriptomics analysis and translomics analysis,the characteristics of gene expression under hypoxic conditions was revealed.We first treated SW620 cells with the compound cobalt chloride to successfully simulate hypoxia conditions.We then used mRNA-seq and Ribo-seq analyses to show that transcription and translation of many genes are affected by hypoxia,and that the efficiency of gene translation in several tumor-related signaling pathways is also regulated by hypoxia.In addition,real-time PCR and Western blot were used to verify the expression of some hypoxic regulated genes.This study elucidates the expression characteristics of tumor-related genes regulated by hypoxia from a new perspective and helps to clarify the pathogenesis of straight colon cancer.
