摘  要:目的探讨清肝益肾祛风复方对高血压肠道菌群失调的干预效应及可能机制。方法将12只5周龄雄性自发性高血压大鼠(SHR)随机分为模型组及清肝益肾祛风复方组各6只,以6只同周龄WKY雄性大鼠为正常组。从6周龄开始,清肝益肾祛风复方组给予清肝益肾祛风复方浓缩液灌胃,模型组和正常组给予等体积纯净水灌胃,均1次/d,持续至20周龄。采集各组大鼠粪便,进行粪便16s rDNA测序及短链脂肪酸气相色谱分析。结果模型组大鼠粪便16s rDNA测序中Ace指数、Chao指数、Shannon指数及双歧杆菌目、双歧杆菌属相对丰度均显著低于正常组(P均<0.05),清肝益肾祛风复方组以上指标均显著高于模型组(P均<0.05);模型组变形菌门及变形菌纲相对丰度均显著高于正常组(P均<0.05),清肝益肾祛风复方组均显著低于模型组(P均<0.05)。模型组大鼠粪便中乙酸、丙酸、异丁酸相对水平均显著低于正常组(P均<0.05),丁酸、戊酸、异戊酸相对水平与正常组比较差异均无统计学意义(P均>0.05);清肝益肾祛风复方组大鼠粪便中乙酸、丁酸相对水平均显著高于模型组(P均<0.05),丙酸、异丁酸、戊酸、异戊酸相对水平与模型组比较差异均无统计学意义(P均>0.05)。结论高血压大鼠存在肠道菌群失调,清肝益肾祛风复方可促进高血压大鼠肠道双歧杆菌的生长,下调变形菌门菌群的丰度及促进肠道中短链脂肪酸乙酸和丁酸的产生,从而改善肠道菌群失调,维持肠道内稳态。Objective It is to explore the intervention effect and possible mechanism of Qinggan Yishen Qufeng Decocion on the intestinal flora imbalance of hypertension.Methods Twelve 5-week-old male spontaneously hypertensive rats(SHR)were randomly divided into a model group and a Qinggan Yishen Qufeng Decoction group,with 6 rats in each group,and 6 WKY male rats of the same age as the normal group.From 6 weeks of age,the Qinggan Yishen Qufeng Decoction group was given the concentrated liquid of Qinggan Yishen Qufeng Decoction by gavage,and the model group and the normal group were given equal volume of purified water by gavage,all once a day,lasted to 20 weeks age.The feces of the rats in each group were collected for 16S rDNA sequencing and short-chain fatty acid gas chromatography analysis.Results The Ace index,Chao index,Shannon index and the relative abundance of the Bifidobacteriales,and Bifidobacterium in the fecal 16s rDNA sequencing of rats in the model group were significantly lower than those in the normal group(all P<0.05),the above indexes of the Qinggan Yishen Qufeng Decoction group were significantly higher than those of the model group(all P<0.05).The relative abundance of the phylum proteobacteria,the class epsilonproteobacteria in the model group were significantly higher than those in the normal group(all P<0.05),the above indexes of the Qinggan Yishen Qufeng Decoction group were significantly lower than those of the model group(all P<0.05).The relative levels of acetic acid,propionic acid,and isobutyric acid in the feces of the rats in the model group were significantly lower than those of the normal group(all P<0.05),there was no significant difference in the relative levels of butyric acid,valeric acid,and isovaleric acid compared with the normal group(all P>0.05);the relative levels of acetic acid and butyric acid in the feces of rats in the Qinggan Yishen Qufeng Decoction group were significantly higher than those in the model group(all P<0.05),there was no significant difference in the relative lev
