摘  要:目的研究藤黄酸对子宫内膜癌Ishikawa细胞生物学行为的影响和机制。方法子宫内膜癌Ishikawa细胞分成4组:对照组、实验组(用0.8μg·mL^(-1)藤黄酸处理)、Anti-miR-NC组(转染inhibitor control,0.8μg·mL^(-1)藤黄酸处理)、Anti-miR-497-5p组(转染miR-497-5p inhibitor,0.8μg·mL^(-1)藤黄酸处理)。用qRT-PCR方法检测miR-497-5p表达,用MTT法检测细胞增殖,用流式细胞术检测凋亡率,Transwell小室检测迁移能力,用蛋白印迹法(Western blot)检测剪切的含半胱氨酸的天冬氨酸蛋白水解酶3、上皮性钙黏附素、神经性钙黏附素、基质金属蛋白酶-9蛋白表达。结果对照组、实验组、Anti-miR-NC组、Anti-miR-497-5p组子宫内膜癌Ishikawa细胞中miR-497-5p表达量分别为1.00±0.10,1.98±0.17,1.96±0.19和0.99±0.12;上述4组的细胞增殖率分别为(100.00±11.70)%,(55.10±8.10)%,(56.20±7.15)%和(87.51±7.64)%;上述4组的细胞凋亡率分别为(4.66±0.88)%,(12.45±1.85)%,(13.05±1.56)%和(8.72±1.10)%;上述4组的迁移细胞数目分别为215.36±18.42,103.69±9.15,108.42±11.50和164.79±13.57。上述指标,实验组与对照组比较,差异均有统计学意义(均P<0.05);Anti-miR-497-5p组与Anti-miR-NC组比较,差异有统计学意义(P<0.05)。结论藤黄酸通过上调miR-497-5p影响子宫内膜癌Ishikawa细胞生物学行为。Objective To study the effect and mechanism of gambogic acid on the biological behavior of endometrial cancer Ishikawa cells. Methods Endometrial cancer Ishikawa cells were divided into control group, experimental group(0.8 μg·mL;gambogic acid treatment), Anti-miR-NC group(transfection inhibitor control, 0.8 μg·mL;gambogic acid treatment), Anti-miR-497-5 p group(transfection miR-497-5 p inhibitor, 0.8 μg·mL;gambogic acid treatment). miR-497-5p expression was detected by qRT-PCR, cell proliferation was detected by MTT, apoptosis rate was detected by flow cytometry, migration was detected by Transwell chamber, Cleaved Caspase-3, E-cadherin, N-cadherin, MMP-9 protein expressionwas detected by Western blot. Results The expression levels of miR-497-5 p in control group, experimental group, Anti-miR-NC group, Anti-miR -497-5 p group of endometrial cancer Ishikawa cells were 1. 00 ± 0. 10,1. 98 ± 0. 17,1. 96 ± 0. 19 and0. 99 ± 0. 12;the cell proliferation rates were( 100. 00 ± 11. 70) %,( 55. 10 ± 8. 10) %,( 56. 20 ± 7. 15) % and( 87. 51 ± 7. 64) %;the apoptosis rates were( 4. 66 ± 0. 88) %,( 12. 45 ± 1. 85) %,( 13. 05 ± 1. 56) % and( 8. 72 ± 1. 10) %;the number of migrating cells were 215. 36 ± 18. 42,103. 69 ± 9. 15,108. 42 ± 11. 50,164. 79 ± 13. 57. Compared between control group experimental group,the difference was significant( P < 0. 05);Compared between the Anti-miR-497-5 p group and Anti-miR-NC group,the difference was significant( P < 0. 05). Conclusion Gambogic acid affected the biological behavior of endometrial cancer Ishikawa cells by up-regulating miR-497-5 p.
