摘  要:葡萄糖含量是淀粉糖化过程的一个重要指标。现有葡萄糖的测定方法在分析效率、抗干扰性、通用性、分析成本等方面不能满足过程分析的需要。研究通过采集不同反应时间下淀粉糖化液样本的近红外光谱,建立校正集样本,结合角度转换方法计算样本光谱与参比光谱向量系列夹角余弦的方差值EE,建立了EE值与葡萄糖含量的关联方程,方程相关系数r为0.9978,建立了糖化液中葡萄糖含量的近红外光谱快速分析方法。对验证集糖化过程溶液样本,依照所建立的方法求取糖化液中葡萄糖含量,与高效液相色谱法的测定值相比,绝对误差为0.09~2.98 mg/mL,相对误差为0.18%~9.06%,模型预测的相对标准偏差为2.27%~5.20%。最后将建立的快速分析方法应用于不同反应温度下糖化反应过程,实现糖化过程中果糖含量的实时测定。该方法在分析效率、抗干扰性、通用性、分析成本等方面可满足生产过程分析的需要,为淀粉液化液糖化过程分析提供参考。Glucose content is an important index of starch saccharification process.The existing methods for the determination of glucose can not meet the needs of process analysis in terms of analysis efficiency,anti-interference,versatility and analysis cost.The near-infrared spectra of starch glycosylation solution samples at different reaction times were collected to establish correction set samples.Combined with the Angle conversion method,the variance value of cosine of the Angle between sample spectrum and the vector series of reference spectrum was calculated,and the correlation equation between EE value and glucose content was established.The correlation coefficient r of the equation was 0.9978.A fast method for the determination of glucose content in saccharating solution by near infrared spectroscopy was established.Compared with the real value determined by HPLC,the absolute error and relative error were between 0.09~2.98 mg/mL,0.18%~9.06%,and the relative standard deviation of the model prediction were between 2.27%~5.20%.Finally,the established rapid analytical method was applied to the saccharification process at different reaction temperatures to realize the real-time determination of fructose content in the saccharification process.The method can meet the needs of production process analysis in terms of analysis efficiency,anti-interference,versatility and analysis cost,and provide reference for the analysis of the saccharification process of starch liquefaction liquid.
