非小细胞肺癌手术前后长链非编码RNA水平变化及其与预后的关系

摘　　要：目的分析非小细胞肺癌(NSCLC)患者手术前后长链非编码RNA(LncRNAs)水平变化及其与预后的相关性。方法选取2017年4月至2018年5月我院收治的NSCLC患者90例(恶性组)、肺良性肿瘤者54例(良性组)、健康体检者30例(对照组),比较三组血清LncRNA MEG3、LncRNA RAB11B-AS1、LncRNA H19水平,分析NSCLC患者手术前后上述指标变化及与预后的关系。结果恶性组血清LncRNA MEG3水平低于良性组及对照组,LncRNA RAB11B-AS1、LncRNA H19水平高于良性组及对照组(P<0.05);NSCLC患者术后血清LncRNA MEG3水平高于术前,血清LncRNA RAB11B-AS1、LncRNA H19水平低于术前(P<0.05);LncRNA MEG3高表达组总生存时间(OS)较LncRNA MEG3低表达组延长,LncRNA RAB11B-AS1高表达组OS较LncRNA RAB11B-AS1低表达组缩短,LncRNA H19高表达组OS较LncRNA H19低表达组缩短(P<0.05);肿瘤大小、远处转移、TNM分期、LncRNA MEG3低表达、LncRNA RAB11B-AS1高表达、LncRNA H19高表达均为影响NSCLC患者预后的独立危险因素(P<0.05)。结论NSCLC患者手术前后LncRNA MEG3、LncRNA RAB11B-AS1、LncRNA H19水平发生变化,且其与预后均有密切关系,有望成为其分子标志物。Objective To analyze the changes in long non-coding RNAs(LncRNAs)levels in patients with non-small cell lung cancer(NSCLC)before and after surgery,and their correlation with prognosis.Methods Ninety NSCLC patients(malignant group),54 patients with benign lung tumors(benign group),and 30 healthy individuals(control group)were enrolled in this study between April 2017 and May 2018.The levels of serum LncRNA MEG3,LncRNA RAB11 B-AS1 and LncRNA H19 were compared among the three groups.The changes in the above indicators in patients with NSCLC before and after surgery,and their correlation with prognosis were analyzed.Results The level of serum LncRNA MEG3 was lower while the levels of LncRNA RAB11 B-AS1 and LncRNA H19 were higher in the malignant group than those in the benign group and the control group(P<0.05).After operation,the level of serum LncRNA MEG3 was increased,while the levels ofserum LncRNA RAB11 B-AS1 and LncRNA H19 were decreased in the NSCLC patients(P<0.05).The overall survival time(OS)of the LncRNA MEG3 high expression group was longer than that of the LncRNA MEG3 low expression group.The OS of the LncRNA RAB11 B-AS1 high expression group was shorter than that of the LncRNA RAB11 BAS1 low expression group.The OS of the LncRNA H19 high expression group was shorter than that of the LncRNA H19 low expression group(P<0.05).COX regression analysis showed that tumor size,distant metastasis,TNM staging,LncRNA MEG3 low expression,LncRNA RAB11 B-AS1 high expression,and LncRNA H19 high expression were independent risk factors affecting the prognosis of patients with NSCLC(P<0.05).Conclusion Changes in the levels of LncRNA MEG3,LncRNA RAB11 B-AS1 and LncRNA H19 can be observed in patients with NSCLC before and after surgery.The indexes are closely related to the prognosis.The results indicate that the three long non-coding RNAs may be molecular markers for the disease.
