摘 要:目的制备南极海茸多糖果冻,研究其对便秘小鼠的润肠通便作用。方法以南极海茸多糖为原料,制备1款酸甜可口的果冻。采用1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生-高效液相色谱法(HPLC)测定果冻中南极海茸多糖的含量。探索了南极海茸多糖果冻对便秘小鼠润肠通便作用:将昆明小鼠随机分为空白组、模型组、阳性对照组、果冻低剂量组和高剂量组。果冻低剂量组和高剂量组小鼠按0.02 mL/g剂量分别灌胃给予添加量为5%南极海茸多糖果冻溶液;阳性对照组按0.02 mL/g剂量灌胃给予乳果糖口服液;空白组和模型组小鼠给予相同剂量的果冻基质,连续灌胃7 d。采用盐酸洛哌丁胺小鼠便秘模型,观察各组小鼠的首粒排黑便时间、5 h内排便粒数、排便质量以及小肠墨汁推进率。结果以岩藻糖表征南极海茸多糖含量为5.06%,测定值与实际添加量相符。实验结果表明,与模型组相比,南极海茸多糖果冻可显著提高小肠墨汁推进率(P<0.05),明显增加便秘小鼠的排便粒数和排便质量(P<0.05),显著缩短便秘小鼠的首粒排黑便时间(P<0.01)。结论成功制备了南极海茸多糖果冻,PMP柱前衍生-HPLC法检测果冻中岩藻糖含量结果准确,专属性好,可用于南极海茸多糖果冻的质量控制。南极海茸多糖果冻具有良好的润肠通便作用。Objective Durvillaea antarctica polysaccharides(DAP)jelly was prepared and conducted to study the laxative effect.Methods Using Durvillaea antarctica polysaccharides as raw materials,a sweet and sour jelly was prepared.The content of Durvillaea antarctica polysaccharides in jelly was determined by 1-phenyl-3-methyl-5-pyrazolone(PMP)pre-column derivatization and high performance liquid chromatography(HPLC)method based on fucose.To study the laxative effect of Durvillaea antarctica polysaccharides jelly,Kunming male mice were randomly divided into blank group,model group,positive control group,jelly low and high dose groups.Mice in jelly low-dose and high-dose groups were intragastrical administered jelly dilution with 5%Durvillaea antarctica polysaccharides at a dose of 0.02 mL/g,respectively.Positive control group was given lactulose oral solution at a dose of 0.02 mL/g.The blank group and model group were given the same amount of jelly matrix.The drug was administered for 7 consecutive days.The constipation mouse model was established by loperamide hydrochloride.The time of the first black stool,the number and weight of defecated grains within 5h,and the propelling rate of intestinal ink in each group were observed.Results Using HPLC method by pre-column derivatization,the characterization rate of fucose was 5.06%.The results showed that the determination value of fucose was in good agreement with the actual addition amount.The results of animal experiments showed that compared with the model group,the number and weight of defecation of constipated mice were significantly increased(P<0.05),the time of the first black stool was shortened(P<0.01),and the ink propelling rate of small intestine was improved(P<0.05).Conclusion The HPLC method by precolumn derivatization was accurate and good specificity,could be used for the quality control of Durvillaea antarctica polysaccharides jelly.Durvillaea antarctica polysaccharides jelly has a good laxative effect.
