摘 要:目的优化诃子总黄酮提取工艺,并探讨其体外抗XOD、抗氧化活性。方法在单因素试验基础上,以乙醇体积分数、液料比、提取温度、提取时间为影响因素,总黄酮提取率为评价指标,响应面法优化提取工艺。测定提取物对XOD的抑制作用及对DPPH自由基、羟基自由基、亚硝酸根离子的清除能力。结果最佳条件为乙醇体积分数61%,液料比16∶1,提取温度69℃,提取时间99 min,总黄酮提取率为5.28%。提取物对XOD、DPPH自由基、羟自由基、亚硝酸根离子的IC_(50)值分别为29.22μg/mL、0.1277 mg/mL、0.5188 mg/mL、42.78μg/mL。结论该方法简便、稳定、可行,可用于提取体外抗XOD、抗氧化活性较强的诃子总黄酮。AIM To optimize the extraction process for total flavonoids from Terminalia chebula Retz.and to explore their in vitro anti-XOD and anti-oxidation activities.METHODS With ethanol concentration,solid-liquid ratio,ultrasonic power and ultrasonic time as influencing factors,extraction rate of total flavonoids as an evaluation index,the extraction process was optimized by response surface method on the basis of single factor test.The extract’inhibitory effect on XOD and scavenging capacities on DPPH free radical,hydroxyl free radical,nitrous acid ion were determined.RESULTS The optimal conditions were determined to be 61%for ethanol concentration,16∶1 for liquid-solid ratio,69℃for extraction temperature,and 99 min for extraction time,the extraction rate of total flavonoids was 5.28%.The IC_(50)values of extract on XOD,DPPH free radical,hydroxyl free radical and nitrous acid ion were 29.22μg/mL,0.1277 mg/mL,0.5188 mg/mL and 42.78μg/mL,respectively.CONCLUSION This simple,stable and feasible method can be used for the extraction of total flavonoids with strong in vitro anti-XOD and anti-oxidation activities from T.chebula.
