摘  要:目的:探讨下调Lnc RNACTC-459F4.3表达对肝癌细胞干性增殖、凋亡的影响.方法:采用CTC-459F4.3shRNA慢病毒质粒转染HepG2肝癌细胞系,分别设CTC-459F4.3敲低组、对照组、空质粒组.采用RT-PCR方法检测CTC-459F4.3shRNA转染效率;Western-Blot检测干细胞调控因子SOX2和OCT4的相对表达量;瘤球成形实验检测肝癌细胞干性增殖能力;流式技术检测肝癌细胞凋亡率.结果:调低CTC-459F4.3表达可介导SOX2和OCT4的相对表达量上调(P<0.001);CTC-459F4.3敲低组与对照组、空质粒组相比,HepG2干性增殖能力显著增强且细胞凋亡率显著下降(P<0.001);对照组与空质粒组相比细胞干性增殖能力和凋亡率无显著差异(P>0.05).结论:调低Lnc RNACTC-459F4.3表达,可促进肝癌细胞的干性增殖、抑制其凋亡.深入研究CTC-459F4.3生物学特性可以为肝癌靶向诊治提供新思路.Objective:To investigate the effect of down-regulation of Lnc RNA CTC-459F4.3 expression on stem proliferation and apoptosis of hepatocellular carcinoma cells.Methods:CTC-459F4.3 shRNA lentivirus plasmid was used to transfect HepG2 hepatoma cell lines.The cells were divided into three groups:CTC-459F4.3 knockout group,control group and empty plasmid group.The transfection efficiency of CTC-459F4.3 shRNA was detected by RT PCR.Western-Blot was used to detect the relative expression of stem cell regulatory factors SOX2 and OCT4.Tumor ball forming test was used to detect the stem proliferation of hepatoma cells.Flow cytometry was used to detect the apoptosis rate of hepatoma cells.Results:The relative expression of SOX2 and OCT4 was increased by down-regulating the expression of CTC-459F4.3.Compared with the control group and the empty plasmid group,the stem proliferation ability of CTC-459F4.3 knockdown group was significantly enhanced and the apoptosis rate was significantly decreased(P<0.001).There was no significant difference in the stem proliferation ability and apoptosis rate between the control group and the empty plasmid group(P<0.05).Conclusion:Down-regulating the expression of Lnc RNA CTC 459F4.3 can promote the stem proliferation and inhibit the apoptosis of hepatocellular carcinoma cells.In-depth study of the biological characteristics of CTC-459F4.3 can provide new ideas for targeted diagnosis and treatment of HCC.
