西洋参片对癫痫模型大鼠海马组织GFAP和Fos表达的影响及机制研究

摘  要:目的探讨西洋参对癫痫模型大鼠海马组织神经胶质纤维酸性蛋白(GFAP)和Fos蛋白表达的影响,并分析其机制。方法40只6周龄左右清洁级Sprague Dawley雄性大鼠根据实验目的分为4组,分别为正常对照组、癫痫模型组、常规组和西洋参组,选择其中3组采用戊四氮腹腔注射建立癫痫模型,其中,正常对照组和癫痫模型组采用等量0.9%氯化钠溶液腹腔注射和灌胃,常规组实施50 mg/kg苯妥英钠腹腔注射,西洋参组在常规组基础上进行3 g/kg西洋参片灌胃,4组均连续治疗1个月。记录各组大鼠癫痫发作等级;分别于治疗前1 d、治疗后10 d以及治疗后1个月,采用水迷宫实验评估各组大鼠学习记忆能力,酶联免疫吸附试验试剂盒检测各组大鼠血清和海马组织中超敏C反应蛋白(hs-CRP)和热休克蛋白70(HSP-70)水平,免疫组织化学检测各组大鼠海马组织GFAP、Fos表达。结果治疗后,癫痫模型组、常规组和西洋参组大鼠癫痫分级间相比差异有统计学意义,西洋参组低等级(Ⅰ~Ⅱ级)癫痫大鼠占比显著高于癫痫模型组,差异有统计学意义(P<0.05)。治疗前1 d~治疗后1个月,常规组和西洋参组大鼠滞留时长和穿过平台次数均随治疗时间延长而显著增加,血清hs-CRP和HSP-70水平均随治疗时间延长而显著降低,差异均有统计学意义(P<0.05)。治疗后1个月,与正常对照组相比,其余3组癫痫大鼠滞留时长及穿过平台次数均显著减少,血清hs-CRP和HSP-70水平、海马组织GFAP和Fos阳性表达评分及hs-CRP和HSP-70水平均显著增加,差异均有统计学意义(P<0.05);与癫痫模型组相比,常规组和西洋参组大鼠滞留时长及穿过平台次数均显著增加,血清hs-CRP和HSP-70水平、海马组织GFAP和Fos阳性表达评分及hs-CRP和HSP-70水平均显著降低,且西洋参组也显著优于常规组,差异均有统计学意义(P<0.05)。结论西洋参片辅助干预可显著改善癫痫模型大鼠癫痫分级以及�Objective To investigate the effect of American ginseng on the expression of glial fibrillary acidic protein(GFAP)and Fos in hippocampus of epilepsy model rats,and analyze its mechanism.Methods A total of 40 clean-grade Sprague Dawley male rats aged about 6 weeks were divided into 4 groups according to the purpose of the experiment,namely the normal control group,the epilepsy model group,the routine group and the American ginseng group.The three groups were selected to use PTZ intraperitoneal injection to establish epilepsy models.Among them,the normal control group and the epilepsy model group were injected with the same amount of normal saline and gavage,and the routine group was injected with 50 mg/kg phenytoin sodium intraperitoneally,and the American ginseng group was given 3 g/kg American ginseng tablets by gavage on the basis of the conventional group,and the four groups were treated continuously for 1 month.The grades of seizures of each group of rats were recorded;1 d before treatment,10 d after treatment,and 1 month after treatment,the learning and memory ability of each group was evaluated by the water maze experiment,the enzyme-linked immunosorbent assay(ELISA)kit detects the levels of high sensitivity C-reactive protein(hs-CRP)and heat shock protein 70(HSP-70)in the serum and hippocampus of each group of rats,and immunohistochemical detection of the expression of GFAP and Fos in the hippocampus of each group of rats.Results After treatment,there was a statistically significant difference in the grades of epilepsy among the Epilepsy model group,routine group and American ginseng group of rats.The proportion of epileptic rats with low grade(gradeⅠ~Ⅱ)in the American ginseng group was significantly higher than that in the epilepsy model group(P<0.05).From 1 d before treatment to 1 month after treatment,the retention time and the number of times the rats in the conventional group and the American ginseng group crossed the platform increased significantly with the treatment time,and the levels of serum
