摘 要:目的研究乳酸杆菌在大肠癌小鼠中对顺铂的抗肿瘤作用的影响,并探讨其机制。方法人结肠癌细胞株SW620根据不同的处理方式将细胞分为3组:对照组、顺铂组和乳酸杆菌+顺铂组。对照组细胞不进行任何处理;顺铂组细胞培养基中添加5.0μmol/L顺铂;乳酸杆菌+顺铂组细胞使用Transwell小室进行培养,下室接种SW620细胞,上室接种乳酸杆菌,培养基添加5.0μmol/L顺铂。分别于处理0、12、24、36、48 h后,采用细胞凋亡检测试剂盒检测细胞凋亡率;处理48 h后,采用Annexin V-FITC细胞凋亡检测试剂盒检测细胞周期。取30只6~8周龄Balb/c小鼠,通过腋下皮下注射人大肠癌细胞系SW620建立大肠癌荷瘤模型,然后按照随机数表法分为对照组、顺铂组和乳酸杆菌+顺铂组。顺铂组小鼠腹腔注射顺铂治疗,乳酸杆菌+顺铂组腹腔注射顺铂和乳酸杆菌灌胃治疗,对照组给予等量的0.9%氯化钠溶液。2周后,安乐死各组小鼠以获取外周血和肿瘤组织,比较肿瘤组织大小及各组小鼠外周血T细胞亚群比率。结果在体外,与顺铂组相比,乳酸杆菌+顺铂组大肠癌细胞凋亡率显著增高,乳酸杆菌+顺铂组大肠癌细胞G0/G1和G2/M期细胞比率显著下降,而S期细胞比率均显著上升,差异均有统计学意义(P<0.05)。在体内,与顺铂组相比,乳酸杆菌+顺铂组大肠癌细胞荷瘤小鼠肿瘤组织重量和体积均显著降低,外周血CD4^(+)细胞比率均显著上升,并且CD4^(+)/CD8^(+)比率均显著下降,差异均有统计学意义(P<0.05)。结论乳酸杆菌可以在体内增强顺铂对大肠癌小鼠的抗肿瘤作用,其机制可能与协同抑制大肠癌细胞的增殖和促进凋亡,以及增强大肠癌小鼠免疫功能有关。Objective To study the effect of Lactobacillus on the anti-tumor effect of cisplatin in mice with colorectal cancer,and to explore its mechanism.Methods The human colon cancer cell line SW620 divides the cells into 3 groups according to different treatment methods:the control group,the cisplatin group and the lactobacillus+cisplatin group.Cells in the control group were not subjected to any treatment;cells in the cisplatin group were added with 5.0μmol/L cisplatin;cells in the Lactobacillus+cisplatin group were cultured in a Transwell chamber.The lower chamber was inoculated with SW620 cells,the upper chamber was inoculated with Lactobacillus,and the medium was supplemented with 5.0μmol/L cisplatin.After 0,12,24,36,and 48 hours of treatment,the apoptosis detection kit was used to detect the apoptosis rate;After 48 hours of treatment,the Annexin V-FITC cell apoptosis detection kit was used to detect the cell cycle.30 Balb/c mice aged 6 to 8 weeks were injected into the armpit of human colorectal cancer cell line SW620 to establish a tumor-bearing colorectal cancer model.Then according to the random number table method,they were divided into control group,cisplatin group and lactobacillus+cisplatin group.The mice in the cisplatin group were treated by intraperitoneal injection of cisplatin,the lactobacillus+cisplatin group was treated by intraperitoneal injection of cisplatin and lactobacillus,and the control group was given the same amount of normal saline.Two weeks later,the mice in each group were euthanized to obtain peripheral blood and tumor tissue,and the tumor tissue size and the proportion of T cell subsets in the peripheral blood of the mice in each group were compared.Results In vitro,compared with the cisplatin group,the apoptotic rate of colorectal cancer cells in the Lactobacillus+cisplatin group was significantly increased,and the proportion of colorectal cancer cells in the G0/G1 and G2/M phases of the Lactobacillus+cisplatin group was significant decreased,while the proportion of S-phase cells wa
