摘 要:目的探讨rmhTNF对Δ133p53表达差异的胃癌裸鼠皮下移植瘤的作用及其机制。方法以人胃癌MKN-45和SGC-7901细胞为研究材料,建立裸鼠皮下移植瘤模型,随机分成3组,空白对照组肌注生理盐水0.1ml,实验1组肌注rmhTNF 16.7IU,实验2组肌注rmhTNF 33.3IU,均为1次/d,连续注射10d。14d后处死裸鼠,剥离肿瘤组织,记录肿瘤体积和抑瘤率,采用NRT-PCR检测Δ133p53mRNA的表达水平。结果rmhTNF对MKN-45胃癌裸鼠移植瘤有抑制作用(F=937.773,P<0.01),而且具有量效及时效依赖关系。而对SGC-7901胃癌裸鼠移植瘤抑制作用不明显(t=-0.121,P>0.05)。两种胃癌组织中,胃癌MKN-45表达Δ133p53基因,而SGC-7901不表达Δ133p53基因;在MKN-45移植性胃癌中,rmhTNF可下调Δ133p53基因,具有量效关系(F=114.265,P<0.01)。结论肌肉注射rmhTNF对表达Δ133p53裸鼠胃癌生长具有显著抑制作用,可能是通过调节Δ133p53的表达实现的。Objective To explore the role ofΔ133p53 in the effect and mechanism of recombined and modified human Tumor Necrosis Factor on subcutaneous xenograft tumor of gastric cancer in nude mice.Methods The subcutaneous xenograft model of nude mice was established with MKN-45 or SGC-7901 cells and were treated with rmhTNF of different concentrations only for 10 days,once a day,and the tumor volume was measured.Thereafter,tumor tissues were collected,and tumor inhibition rate was calculated 14 days after injection.mRNA ofΔ133p53 was measured by nested reverse transcription PCR.Results On MKN-45 xenograft tumor with positiveΔ133p53 expression,the effect of rmhTNF was significant in growth inhibition test with remarkable time-effect and dose-effect(F=937.773,P<0.01).However,on SGC-7901 xenograft tumor with negativeΔ133p53 expression,no growth inhibition was showed by rmhTNF(t=-0.121,P>0.05).In mRNA measurement,down-regulation ofΔ133p53 was significant in MKN45 xenograft tumor treated by rmhTNF with remarkable dose-effect(F=114.265,P<0.01).Conclusion InΔ133p53 positively expressed MKN45 xenograft tumor of nude mice,rmhTNF was showed an effective tumor inhibitor,and this effect might be helped by down-regulation ofΔ133p53.
