摘 要:目的探讨不同强度运动对高脂饮食大鼠窦房结及心功能的影响。方法将24只8周龄雄性Sprague Dawley大鼠随机分为A、B、C、D组,每组6只。A组大鼠给予普通饲料饲养,B组大鼠给予高脂饲料饲养,2组大鼠均笼内饲养不运动;C组大鼠给予高脂饲料饲养,同时进行8周无负重游泳运动,每日60 min,每周6 d;D组大鼠给予高脂饲料饲养,同时进行8周高强度(5%体质量负重)游泳运动,每日60 min,每周6 d。末次游泳训练24 h后,应用彩色多普勒超声诊疗仪测试大鼠左心室收缩末期内径(LVDS)、左心室舒张末期内径(LVDD)、左心室收缩末期容积(LVVS)、每搏输出量(SV)、左心室射血分数(LVEF)、左心室缩短分数(LVSF)等心功能指标;大鼠用体积分数10%水合氯醛麻醉后,开胸取窦房结,采用苏木精-伊红染色观察窦房结形态结构变化,油红O染色法测定窦房结脂肪细胞分布及脂肪细胞阳性表达量,免疫组织化学法测定窦房结神经标志物蛋白基因产物9.5(PGP 9.5)表达。结果B组大鼠LVDS、LVDD、LVVS显著高于A组(P<0.05),SV、LVEF、LVSF显著低于A组(P<0.05)。C组大鼠LVDS、LVDD、LVVS、SV、LVEF与A组比较差异无统计学意义(P>0.05),LVSF显著高于A组(P<0.05)。C组大鼠LVDS、LVDD、LVVS显著低于B组,SV、LVEF、LVSF显著高于B组(P<0.05)。D组大鼠SV显著低于C组,LVSF显著高于A组,LVEF、LVSF显著高于B组,SV显著低于C组(P<0.05);D组大鼠其余心功能指标分别与A组、B组及C组比较差异均无统计学意义(P>0.05)。与A组大鼠比较,B组大鼠窦房结结构缺失,细胞间隙明显增宽、肌纤维排列混乱;C组大鼠窦房结形态结构良好,细胞间隙均匀较窄,细胞质染色均匀;与C组相比,D组大鼠窦房结形态结构欠佳,细胞间隙略增宽。B组、C组、D组大鼠窦房结区脂肪细胞阳性表达量显著高于A组,B组大鼠窦房结区脂肪细胞阳性表达量显著高于C组和D组,D组大鼠窦房结区脂肪细胞阳性表达量�Objective To investigate the protective effect of different intensity exercises on sinus node and cardiac function in rats fed with a high-fat diet.Methods Twenty-four 8-week-old male SD rats were randomly divided into group A,group B,group C and group D,with 6 rats in each group.The rats in group A were fed with normal diet,the rats in group B were fed with high-fat diet,and rats in the both groups were kept in cages without exercise;the rats in group C were fed with high-fat diet and performed with swimming without weight-bearing for eight weeks,60 minutes once day for six days in one week;the rats in group D were fed with high-fat diet and performed with high intensity swimming(5%body weight-bearing)for 8 weeks,60 minutes once day for 6 days once week.At 24 hours after the last swimming training,the cardiac function indexes including left ventricular end systolic internal meridian(LVDS),left ventricular end diastolic internal meridian(LVDD),left ventricular end systolic volume(LVVS),stroke volume(SV),Left ventricular ejection fraction(LVEF)and left ventricular shortening fraction(LVSF)were measured by color Doppler ultrasound under;After anesthetized with 10%chloral hydrate,the rats were performed with thoracotomy,and sinoatrial node was obtained,the morphological and structural changes of sinoatrial node were observed by hematoxylin-eosin staining,the distribution and positive expression of adipocytes in sinoatrial node were measured by oil red O staining,the expression of nerve marker protein gene product 9.5(PGP 9.5)in sinoatrial node was measured by immunohistochemistry.Results The LVDS,LVDD and LVVS of rats in group B were significantly higher than those in group A,the SV,LVEF and LVSF of rats were significantly lower than those in group A(P<0.05).There was no significant difference in LVDS,LVDD,LVVS,SV and LVEF of rats between the group C and group A(P>0.05),and LVSF was significantly higher than that in group A(P<0.05).The LVDS,LVDD and LVVS of rats in the group C were significantly lower than those in
