摘 要:目的探究黑果小檗总花色苷对帕金森大鼠认知行为能力、神经保护及Klotho基因表达的影响。方法将SPF级雄性大鼠随机分为模型组、假手术组、低剂量组、中剂量组和高剂量组,每组15只。比较各组大鼠神经行为学评分、认知和学习记忆能力、神经元凋亡和Klotho、Foxo3a、p-Foxo3a蛋白表达。结果与假手术组相比,模型组旋转圈数及神经元凋亡增加,悬挂实验评分和PT%、T%值降低(均P<0.05);与模型组相比,低、中、高剂量组旋转圈数及神经元凋亡减少,悬挂实验评分和PT%、T%值升高(均P<0.05)。与假手术组相比,模型组Klotho和Foxo3a蛋白表达减少,p-Foxo3a蛋白表达增加(均P<0.05);与模型组相比,低、中、高剂量组Klotho和Foxo3a蛋白表达增加,p-Foxo3a蛋白表达减少(均P<0.05)。结论黑果小檗总花色苷可改善帕金森大鼠认知行为能力,保护PD大鼠的神经功能,其作用机制可能是通过调控Klotho、Foxo3a信号通路实现。Objective To investigate the effects of Berberis heteropoda Schrenk total anthocyanins(BHSTA)on cognitive behavior,neuroprotection and KLOTHO gene expression in Parkinson’s rats.Methods Parkinson’s rat models were randomly divided into control group,model group(Parkinson’s disease rat model group),low dose group(Parkinson’s disease rat model given low doses of BHSTA),medium dose group(Parkinson’s disease rat model given medium doses of BHSTA)and high dose group(Parkinson’s disease rat model given high doses of BHSTA),15 in each group.Neurological score and cognitive function of rats were measured.The oxidative stress indexes(SOD,MDA and GSH-Px)of rats in each group were detected.The neuronal apoptosis of rats in each group was detected by TUNEL method.The protein expressions of Klotho,FoxO3 a and p-FoxO3 a in each group were detected(Western blot).The m RNA expressions of Klotho and FoxO3 a in each group were detected(RT-PCR).Results Compared with sham operation group,the number of turns of rotation in model group was significantly increased,and the score of hanging experiment was significantly decreased(bothP<0.05).Compared with model group,the numbers of turns of rotation in low-dose,medium-dose and high-dose groups were significantly decreased,and the scores of hanging experiment were significantly increased(bothP<0.05).The protein and m RNA expressions of Klotho and FoxO3 a in model group were significantly decreased compared with sham operation group,while the protein expression of p-FoxO3 a was significantly increased compared with sham operation group(allP<0.05).The protein and mRNA expressions of Klotho and FoxO3 a in low-dose,medium-dose and high-dose groups were increased in different degrees compared with model group,while the protein expression of p-FoxO3 a was significantly decreased compared with model group(allP<0.05).Conclusions The Berberis heteropoda Schrenk total anthocyanins can improve the cognitive and behavioral ability,protect the neural function of Parkinson’s rats,and promo
