摘  要:目的探讨BNP基因启动子甲基化与风湿性心脏病继发性肺动脉高压发生机制之间的关系。方法收集肺动脉高压病例组和正常对照组外周静脉血样,提取血浆DNA,通过甲基化修饰检测BNP基因位点甲基化水平,并用焦磷酸测序验证结果,以期找出相关基因位点甲基化水平与肺动脉高压之间的关联。结果BNP基因pos.1~5五个位点区域的Cp G岛甲基化率与正常对照组相比,pos.2、pos.4和pos.53个位点的Cp G岛甲基化与正常组差异均有统计学意义(均P<0.05)。结论肺动脉高压患者BNP基因甲基化表达水平有差异,可为临床提供相关参考。Objective To investigate the relationship between the methylation of brain natriuretic peptide(BNP)gene promoter and the pathogenesis of secondary pulmonary hypertension in rheumatic heart disease.Methods Peripheral venous blood samples were collected from pulmonary hypertension patients and normal controls,and plasma DNA was extracted to detect the methylation level of BNP gene sites through methylation modification,and then,the results were verified by pyrosequencing in order to find out the association between the methylation level of relevant gene sites and pulmonary hypertension.Results The methylation rate of CpG island of pos.1~5 sites of BNP gene was compared with that of the normal control group,and there was a significant difference between the methylation of Cp G at pos.2,pos.4 and pos.5 sites and the normal group(P<0.05).Conclusions BNP gene methylation expression is different in patients with pulmonary hypertension,which can provide relevant reference for clinical practice.
