摘  要:以浙江引进的16份迷你甘薯主推品种及潜力品系为对象,采用形态学性状聚类分析和ISSR荧光标记毛细管电泳技术进行遗传多样性分析。结果表明,基于21项形态学性状的聚类图,在欧式距离为8.69处可分为两大类群,类群Ⅰ:浙13、浙33和QZ 66;类群Ⅱ为:浙259、S 8、浙20、浙15、浙132、浙255、心玉、心香、红香蕉、浙6025、浙70、浙75和JS-84。ISSR分子标记分析表明,利用6对引物共进行PCR扩增,迷你甘薯种质资源的观测等位基因(Na)为3个,平均有效等位基因数(Ne)为2.3517个,期望杂合度(He)为0.2186,香农指数(I)为0.6346,通过NTSYSpc 2.1软件进行聚类分析,在遗传距离为1.05时,16份供试迷你甘薯材料可分为两大类。部分供试材料的ISSR标记聚类结果与形态学标记在遗传背景和类群划分上具有一致性,但是两种鉴定分类方法的聚类分析结果也存在一定差异。迷你甘薯种质材料遗传多样性丰富,将形态学及ISSR标记结合能有效提高品种间特异性的鉴定,更能客观、准确地确定种质资源的亲缘关系。In this paper,genetic diversity of 16 mini sweet potato varieties and potential lines introduced from Zhejiang province were analyzed by morphological clustering analysis and ISSR fluorescence labeled capillary electrophoresis technique.The results showed that these germplasms could be divided into two groups at the Euclidean distance of 8.69 according to cluster map based on 21 morphological traits.Among them,the group 1 contained Zhejiang 13,Zhejiang 33 and QZ 66,while the group 2 incorporated Zhe 259,S 8,Zhe 20,Zhe 15,Zhe 132,Zhe 255,Xinyu,Xinxiang,Hongxiangjiao,Zhe 6025,Zhe 70,Zhe 75 and JS-84.Simultaneously,ISSR molecular marker analysis by PCR amplification with 6 pairs of primers showed that there were 3 observed alleles(Na)and the average number of effective alleles(Ne)was 2.3517.And,the expected heterozygosity(He)and the Shannon index(I)were 0.2186 and 0.6346 respectively.Meanwhile,cluster analysis was carried out by NTSYSpc 2.1 software,and the 16 tested mini sweet potato materials could be divided into two categories when the genetic distance was 1.05.ISSR marker clustering results of some tested materials were consistent with morphological markers in terms of genetic background and group division,but there were some differences in clustering analysis results of the two identification and classification methods.In conclusion,the combination of morphology and ISSR markers could effectively improve the identification of specificity between varieties and determine the genetic relationship of germplasm resources objectively and accurately.
