摘 要:目的:观察去甲斑蝥酸钠对恶性黑色素瘤A375细胞的抑制作用,并探讨其作用机制。方法:以不同浓度(6.25、12.5、25、50ug/mL)去甲斑蝥酸钠作用于A375细胞,分别培养24h、48h、72h,观察药物对细胞增殖的影响。药物作用48h后检测细胞凋亡率及凋亡蛋白caspase-3表达变化,并检测药物作用24h细胞周期改变。结果:CCK-8结果显示去甲斑蝥酸钠能够显著抑制A375细胞的增殖,并呈现浓度依赖性和时间依赖性。药物作用48h后25ug/mL、50 ug/mL两组细胞凋亡率明显增加,显著高于对照组(P<0.05),分别为23.58%和45.26%。凋亡蛋白caspase-3表达量也随药物浓度的增加而相应升高。细胞周期检测显示25ug/mL、50 ug/mL药物处理组A375细胞G_(2)/M期延长,G_(0)/G_(1)期缩短,具有统计学差异(P<0.05)。结论:去甲斑蝥酸钠能够剂量及时间依赖性地抑制恶性黑色素瘤A375细胞的增殖,该作用与诱导细胞凋亡以及细胞周期G_(2)/M期阻滞有关。Objective:It aims to investigate the effect of sodium norcantharidate of malignant melanoma A375 cells and its mechanisms.Methods:The A375 cells were treated with sodium norcantharidate at different concentrations(6.25,12.5,25,50ug/mL)for 24 h,48 h and 72 h,respectively.The apoptosis rate and the expression of caspase-3 were determinated after 48 hours of drug treatment,and the cell cycle was measured after 24 hours of drug treatment.Results:CCK-8 results indicated that the proliferation of A375 cells were significantly inhibited by sodium norcantharidate with obvious time-and dosage-dependent.The apoptosis rate of 25 ug/mL and 50 ug/mL groups were 23.58%and 45.26%respectively after 48 h treatment,which were significantly higher than that of the control group(P<0.05).Meanwhile,the protein expression of caspase-3 increased significantly.Cell cycle analysis showed that the G_(2)/M phase was prolonged and the G_(0)/G_(1) phase was shortened in A375 cells treated with 25ug/mL and 50ug/mL sodium norcantharidate.Conclusions:Sodium Norcantharidin could inhibit the proliferation of malignant melanoma A375 cells in a time-and dose-dependent manner,which is related to the induction of apoptosis and the arrest of G_(2)/M cycle.
