摘  要:目的探讨六君子汤对重症急性胰腺炎(SAP)大鼠肠道损伤和蛋白激酶B(Akt)/核因子(NF)-κB信号的影响。方法大鼠分为对照组、SAP组、治疗1组(SAP模型,4 ml/kg六君子汤治疗)、治疗2组(SAP模型,8 ml/kg六君子汤治疗)、治疗3组(SAP模型,16 ml/kg六君子汤治疗)、阳性组(SAP模型,地塞米松治疗),微量法检测血清中淀粉酶水平,可见分光光度法检测血清脂肪酶水平,用肌氨酸氧化酶法检测血清肌酐水平,酶联免疫吸附试验(ELISA)检测血清D-乳酸、内毒素、肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6、IL-1β水平,比色法检测血清中二胺氧化酶(DAO)水平,计算胰腺比重和腹水量,苏木素-伊红(HE)染色对胰腺和肠道组织病理学情况进行评分,Western印迹检测肠道组织中NF-κB/Akt信号通路相关蛋白表达。结果与对照组比,SAP组血清淀粉酶、脂肪酶、肌酐、D-乳酸、DAO、内毒素、TNF-α、IL-6、IL-1β水平均升高,大鼠腹水量、胰腺组织病理评分、肠道组织病理评分和胰腺比重均升高,肠道组织中NF-κBp65、p-Akt/Akt表达量均升高,差异有统计学意义(P<0.05)。与SAP组比,治疗1组、治疗2组、治疗3组、阳性组血清淀粉酶、脂肪酶、肌酐、D-乳酸、DAO、内毒素、TNF-α、IL-6、IL-1β水平均降低,大鼠腹水量、胰腺组织病理评分、肠道组织病理评分和胰腺比重均降低,肠道组织中NF-κBp65、p-Akt/Akt表达量均减少,且治疗1组、治疗2组、治疗3组血清淀粉酶、脂肪酶、肌酐、D-乳酸、DAO、内毒素、TNF-α、IL-6、IL-1β水平均呈逐渐下降趋势,大鼠腹水量、胰腺组织病理评分、肠道组织病理评分和胰腺比重均呈逐渐下降趋势,肠道组织中NF-κBp65、p-Akt/Akt表达量均呈逐渐下降趋势,差异有统计学意义(P<0.05)。结论六君子汤改善SAP大鼠肠道损伤,抑制Akt/NF-κB信号激活。Objective To investigate the effects of Liujunzi decoction on intestinal injury and Akt/NF-κB signals in rats with severe acute pancreatitis.Methods Rats were divided into control group,severe acute pancreatitis(SAP)group,treatment group 1(severe acute pancreatitis model,4 ml/kg Liujunzi decoction treatment),and treatment group 2(severe acute pancreatitis model,8 ml/kg Liujunzi decoction treatment),treatment group 3(severe acute pancreatitis model,16 mL/kg Liujunzi decoction treatment),positive group(severe acute pancreatitis model,dexamethasone treatment),the serum amylase was detected by a micro method level,visible spectrophotometry was used to detect serum lipase level,sarcosine oxidase method was used to detect serum creatinine level,enzyme-linked immunosorbent assay(ELISA)method was used to detect serum D-lactic acid,endotoxin,tumor necrosis factor(TNF)-α,interleukin(IL)-6,IL-1βlevels,colorimetric method was used to detect the level of diamine oxidase(DAO)in the serum,calculation of the specific gravity of the pancreas and ascites volume,HE staining was used to score the pathology of the pancreas and intestines,Western blot method was used to detect the nuclear factor(NF)-κB/protein kinase B(Akt)signal pathway related protein expression in intestinal tissues.Results Compared with the control group,the serum levels of amylase,lipase,creatinine,D-lactic acid,DAO,endotoxin,TNF-α,IL-6,IL-1βin the SAP group increased,and the amount of ascites in the rats increased,pancreatic histopathological score,intestinal histopathological score,and pancreas specific gravity all increased,and the expression levels of NF-κBp65 and p-Akt/Akt in intestinal tissues all increased(P<0.05).Compared with the SAP group,there were amylase,lipase,creatinine,D-lactic acid,DAO,endotoxin,TNF-α,IL-6,IL-1βin the serum of rats in treatment group 1,treatment group 2,treatment group 3 and positive group decreased,rat ascites volume,pancreatic histopathological score,intestinal histopathological score and pancreas specific gravity all
