摘  要:为建立一个简捷、经济、准确可靠的杂交冬瓜种子纯度鉴定方法,通过对‘墨地龙’冬瓜父母本进行全基因组重测序,开发差异显著的InDel标记;以‘墨地龙’杂交种及其父母本为供试DNA,利用开发的InDel分子标记对‘墨地龙’冬瓜杂交种子进行纯度鉴定,同时和田间鉴定结果进行对比。通过基因组重测序筛选出‘墨地龙’冬瓜亲本差异InDel标记共466对,选择其中差异较大的26条进行扩增,发现均可扩增出条带,其中7对引物可明显区分冬瓜样品纯度。InDel分子标记鉴定的纯度与田间小区种植鉴定结果的吻合度达99%以上,表明不同方法获得‘墨地龙’纯度结果高度一致。本研究筛选的In Del分子标记可用于‘墨地龙’冬瓜杂交种的纯度鉴定。In order to establish a simple,economic,accurate and reliable method to identify the purity of wax gourd hybrid seeds,an InDel marker characterized by significant differences was developed via whole genome re-sequencing of parent plants of’Modilong’wax gourd;then the developed In Del molecular marker was used to identify the purity of hybrid seeds of’Modilong’taking the DNA of its hybrid seeds and parent plants as test DNA,and the obtained results were compared with field identification results.A total of 466 pairs of InDel markers were screened by genome resequencing of parent plants of’Modilong’,and 26 of which with distinct differences were selected to be amplified and both got the strip.Among them,7 pairs of primers can clearly distinguish the purity of wax gourd samples.The purity of InDel molecular marker identification was more than 99%consistent with the results of field plot planting identification,indicating that the purity results obtained by different methods were highly consistent.The InDel molecular marker screened in this study could be used as the purity identification of’Modilong’wax gourd hybrids.
