摘 要:目的:研究核受体辅激活蛋白7(NCOA7)在缺氧条件下对口腔鳞癌细胞糖酵解过程的调节作用。方法:采用免疫荧光双染的方法检测口腔鳞癌组织中NCOA7和芳香烃受体(AhR)的表达。用氯化钴构建人舌鳞癌细胞株(SCC9)的缺氧模型。通过siRNA转染、RT-PCR和蛋白印迹等方法分析NCOA7与AhR的相互调节作用及对细胞糖酵解水平的影响。结果:NCOA7和AhR在口腔鳞癌组织细胞核中表达均高于癌旁组织(P<0.0001)。在缺氧条件下,NCOA7的沉默增强了SCC9细胞的糖酵解水平(P<0.05),而激活AhR可以部分逆转这一过程(P<0.05)。结论:NCOA7在缺氧条件下可通过AhR干预口腔鳞癌的糖酵解水平。Objective:To investigate the regulation of nuclear receptor coactivator 7(NCOA7)on the glycolysis of oral squamous cell carcinoma cells under hypoxia.Methods:Immunofluorescence double staining was used to detect the expression of NCOA7 and Aryl hydrocarbon receptor(AhR)in OSCC and paracancerous tissues.A hypoxia model of human tongue squamous cell carcinoma cell line(SCC9)was established with cobalt chloride.siRNA transfection,RT-PCR and Western blotting were used to analyze the correlation between NCOA7 and AhR and its effect on glycolysis.Results:NCOA7 and AhR expression was significantly higher in OSCC tissue than that in paracancerous tissue(P<0.0001).In anoxic environment,knocking down of NCOA7 aggravated the glycolysis level of SCC9 cells and activation of AhR could partly reverse this process(P<0.05).Conclusion:NCOA7 can intervene the glycolysis level of OSCC cells by targeting AhR under hypoxia.
