摘 要:目的探讨磷脂酶A2(Lp-PLA2)与冠状动脉粥样硬化大鼠疾病严重程度和稳定性的关系。方法SPF级雄性SD大鼠随机分为健康组、模型1组、模型2组。健康组给予普通饲料喂养并注射等量生理盐水;模型1组连续4 d腹腔注射维生素D320万U/(kg·d),并使用高脂饲料喂养7周;模型2组给予一次性腹腔注射维生素D380万U/kg,喂养方式、时间同模型1组。模型1组和模型2组分别在造模成功后使用阿托伐他汀灌胃4周;健康组大鼠给予等量生理盐水灌胃。记录各组大鼠体质量变化;测量各组大鼠主动脉粥样硬化斑块面积(PA)、血管管腔面积(LA),计算校正斑块面积(PA/LA);检测各组大鼠三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)及Lp-PLA2和超敏C反应蛋白(hs-CRP)水平。结果与健康组比较,模型1、2组大鼠喂养2、3周体质量增加(P<0.05)。模型1、2组大鼠造模后及治疗后血管LA低于健康组(P<0.05)。与造模后比较,模型1、2组治疗后血管LA增加,主动脉粥样硬化PA及PA/LA降低,但模型2组治疗后血管LA低于模型1组,主动脉粥样硬化PA及PA/LA高于模型1组(P<0.05)。与健康组比较,治疗后模型1、2组大鼠TG、TC、LDL-C、hs-CRP含量及Lp-PLA2活性均显著升高,且模型2组TG、TC、LDL-C、Lp-PLA2活性高于模型1组,但hs-CRP含量低于模型1组(P<0.05);与造模后比较,模型1、2组大鼠治疗后TG、TC、LDL-C、hs-CRP含量及Lp-PLA2活性均显著降低(P<0.05)。结论血清Lp-PLA2水平能反映冠状动脉粥样硬化疾病严重程度和稳定性,且随着冠状动脉粥样硬化病变严重程度和不稳定性增加而升高。Objective To investigate correlations between phospholipase A2(Lp-PLA2)with disease severity and stability of coronary atherosclerosis(CAS)in rats.Methods SPF male SD rats were randomly divided into healthy group,model group 1 and model group 2.The healthy group was fed with normal diet and injected with the same volume of physiological saline;model group 1 was given intraperitoneal injection of Vitamin D3[200,000 U/(kg)/d each time]for 4 d consecutively,and the rats were fed with high-fat diet for 7 weeks;model group 2 was given one-time intraperitoneal injection of Vitamin D3[800,000 U/(kg)],and the feeding method and time were the same with model group 1.Model group 1 and 2 were given Atorvastatin intragastrically for 4 weeks after successful modeling,while healthy rats were given the same volume of physiological saline by gavage.Body mass changes of rats were recorded in each group.Values of aortic atherosclerosis plaque area(PA)and vascular lumen area(LA)were measured,and the corrected plaque areas(PA/LA)were calculated,and levels of triglyceride(TG),total cholesterol(TC),low density lipoprotein-cholesterol(LDL-C),Lp-PLA2 and hypersensitive C-reactive protein(hs-CRP)were detected among all groups.Results Compared with those in healthy group,values of body weight of rats in model group 1 and 2 were significantly increased after feeding for 2 and 3 weeks(P<0.05).LA areas of rats in model group 1 and 2 after modeling and treatment were significantly lower than those in healthy group(P<0.05).Compared with those after modeling,LA areas were significantly increased,while values of aortic atherosclerosis PA and PA/LA were significantly decreased after treatment in model group 1 and 2;compared with those in model group 1,LA value was significantly lower,while values of PA area and PA/LA were significantly higher after treatment in model group 2(P<0.05).Compared with those in healthy group,levels of TG,TC,LDL-C and hs-CRP and the activity of Lp-PLA2 were significantly increased after treatment in model group 1 and 2;
