摘 要:筛选优良根际促生菌株,定位其关键功能基因,对菌株的改造及实际应用具有重要意义。本文以解盐促生菌Enterobacter hormaechei Wu-15为材料,采用第二代和第三代测序技术对该菌株进行全基因组测序,并对其解盐促生相关的基因进行解析。结果表明,植物根际促生菌Wu-15基因组全长为4614006 bp,GC含量55.80%,含有4240个蛋白编码基因;GO、COG和KEGG数据库注释结果表明基因组中含有多种解盐促生相关基因;通过比对分析,筛选出与解盐促生功能相关的基因(溶磷相关基因7个、渗透调节物质相关基因14个、逆向转运蛋白相关基因6个、IAA合成相关基因ipdC、ACC脱氨酶相关基因dcyD)。本研究通过基因筛选及分析,从分子水平证明该菌株解盐促生的能力,为菌株的进一步优化改良提供指导,为该菌株在农业生产中的应用奠定基础。Screening excellent rhizosphere growth-promoting strains and locating their key functional genes are of great significance to the transformation and practical application of strains.In this paper,the salt-reducing and growth-promoting bacteria Enterobacter hormaechei Wu-15 was used as the material,and the second and third-generation sequencing technologies were used to sequence the whole genome of this strain,and the genes related to salt-reducing and growth-promoting were analyzed.The results showed that the total length of plant rhizosphere growth-promoting bacteria Wu-15 genome was 4614006 bp and the GC content was 55.80%,contains a total of 4240 genes;The results of GO,eggNOG and KEGG database annotations show that the genome contains a variety of salt-releasing and growth-promoting genes.Through comparison and analysis,the genes related to salt-dissolving and growth-promoting functions were discovered(7 phosphate-dissolving related genes,14 osmotic regulating substance related genes,6 antiporter related genes,IAA synthesis related genes ipdC,ACC deaminase related gene dcyD).Through genetic screening and analysis,this study proved the ability of the strain to dissolve salt and promote growth at the molecular level,provide guidance for the further optimization and improvement of the strain,and lay the foundation for the application of the strain in agricultural production.
