摘 要:目的报告C57BL/6J小鼠与豚鼠圆窗电诱发听性脑干反应(electrical evoked auditory brainstem response,EABR)波形特点并作比较分析。方法选取16只8周健康的雄性C57BL/6J小鼠及13只健康的白色红目豚鼠。吸入麻醉后,均左耳开放听泡,刺激电极正极置于圆窗龛,刺激电极负极置于同侧乳突皮下,记录电极置于两外耳道口连线与颅正中线交叉处皮下,参考电极置于对侧乳突皮下,地极置于鼻尖皮下。在电刺激条件下通过体表电极记录EABR,比较二者圆窗EABR阈值、潜伏期和幅值的变化及输入输出函数曲线(I/O)。结果C57BL/6J小鼠和豚鼠分别记录到分化良好且稳定的以Ⅱ波或者Ⅲ波为主的EABR波形,C57BL/6J小鼠阈值为0.28±0.07 mA,豚鼠阈值为0.22±0.07 mA,二者间差异没有统计学意义(P>0.05);给予0.8 mA刺激强度时,C57BL/6J小鼠Ⅱ、Ⅲ波潜伏期分别为1.44±0.25 ms、3.18±0.42 ms,豚鼠Ⅱ波、Ⅲ波潜伏期分别为0.82±0.16 ms、2.05±0.26 ms,二者间差异均具有统计学意义(P<0.05);豚鼠潜伏期和幅值变异程度较小,但C57BL/6J小鼠潜伏期及幅值随电刺激量的增加,变化速度较快。结论全麻下C57BL/6J小鼠与豚鼠在蜗外刺激模式下均可以记录到较稳定的EABR波形;豚鼠对电刺激较稳定,而C57BL/6J小鼠对电刺激量的变化较敏感,为听力研究中实验动物的选择提供依据。Objective To report characteristics of round window stimulation EABR in C57BL/6J mice and guinea pigs and comparative analysis.Methods Sixteen male C57BL/6J mice at 8 weeks of age and thirteen male white and red eye guinea pigs were used.The otic vesicle was opened after inhalation anesthesia on the left ear.The anodal stimulating electrode was placed at the round window,and the cathodal stimulating electrode under the ipsilateral mastoid skin.The recording electrode was placed subcutaneously at the intersection of the external auditory meatus plane and the skull mid-line,the reference electrode under the skin of contralateral mastoid and the ground electrode at the nasal tip.The changes of EABR threshold,latency,amplitude and the input/output function curves(I/O)of the two species were analyzed and compared.Results Well-differentiated EABR waveⅡand waveⅢwere recorded from C57BL/6J mice and guinea pigs with an average threshold of 0.28±0.07 mA and 0.22±0.07 mA,and there was no significant difference(P˃0.05).When stimulus intensity was 0.8 mA,the latency of waveⅡand waveⅢin C57BL/6J mice were 1.44±0.25 ms,3.18±0.42 ms respectively,and the latency of waveⅡand waveⅢin guinea pigs were 0.82±0.16 ms,2.05±0.26 ms respectively.The latency of waveⅡand waveⅢwere significantly different between two species(P˂0.05).The degree of variation of the latency and amplitude was small in guinea pigs,but the latency and amplitude of C57BL/6J mice changed faster and the degree of variation was larger with the increase of stimulus intensity.Conclusion Out of cochlea stimulation EABR can be consistently recorded in C57BL/6J mice and guinea pigs under general anesthesia.Guinea pigs were relatively stable to electrical stimulation,C57BL/6J mice was more sensitive to the changes of stimulus intensity,which provided a basis for the selection of experimental animals in future auditory research.
