摘 要:采用紫外光谱、荧光光谱和圆二色谱以及分子对接技术,研究木犀草素(LUTE)与人免疫球蛋白G(hIgG)之间的相互作用。结果表明:LUTE对hIgG有较强的荧光猝灭作用,猝灭机理是形成复合物的静态猝灭,在形成LUTE/hIgG复合物时LUTE更靠近hIgG分子中的色氨酸残基。紫外光谱、三维荧光光谱及圆二色光谱实验结果发现,LUTE与hIgG结合时对蛋白分子中氨基酸残基的微环境影响较大,而对hIgG分子的骨架影响较小。分子模拟与荧光猝灭实验结果说明,二者结合是多种作用力综合作用的结果。The interaction between luteolin(LUTE)and human immunoglobulin G(hIgG)was studied by ultraviolet spectra(UV),fluorescence and circular dichroism(CD)spectra.The results suggested that LUTE had a strong ability to quench the hIgG fluorescence in a static mode.And LUTE was closer to the tryptophan residue in the hIgG molecule when LUTE and hIgG complexed.The results of UV,3D fluorescence and CD showed that LUTE had a great influence on the microenvironment of amino acid residues in hIgG protein molecules,but had little influence on the skeleton of hIgG.The results of molecular simulation and fluorescence quenching experiments showed that the combination of LUTE and hIgG was the result of a variety of forces.
