肿瘤标记物microRNA-21在胰腺癌中的表达及意义

摘  要:目的研究胰腺癌(pancreatic cancer,PC)患者循环microRNA-21(miR-21)的表达水平,并将其与PC相关的临床参数进行相关性分析,以评估其在该病中的临床意义。方法选取2017年1月至2019年1月收治确诊的53例PC患者血清,采用荧光定量多聚核苷酸链式反应(quantitative real-time polymerase chain reaction,QR-PCR)检测miR-21在癌组织、癌旁组织中的表达,并分析其与芽状RTK信号拮抗剂-2(sprouty RTK signaling antagonist-2,SPRY-2)和星型细胞上调基因-1(astrocyte elevated gene-1,AEG-1)等临床参数表达的相关性。结果PC患者癌组织(浸润、转移癌组织及不典型增生组织)的miR-21表达水平显著高于癌旁组织(P<0.05)。PANC-1细胞增殖能力上调与miR-21表达上调呈正比(P<0.05),MIAPaCa-2细胞增殖能力下降时,miR-21表达也随之下调(P<0.05)。PC侵袭能力受DPI的抑制效果与药物浓度呈正比(P<0.05),PC肿瘤细胞转移能力受DPI的抑制效果与药物治疗同样呈正比(P<0.05)。DPI给药是可以明显降低PC患者原代肿瘤细胞内的ROS表达能力(P<0.05),且利用Mito-TEMP 0可阻断四甲基哌啶表达(P<0.05)。Spearman秩相关分析表明,PC患者癌组织中的SPRY-2蛋白表达与miR-21呈负相关(r=-0.381,P<0.05),AEG-1蛋白表达与miR-21无相关性(r=-0.315,P<0.05)。结论PC患癌组织中的miR-21表达水平呈显著的高表达状态,可将其作为PC诊断的一种新型标志物。Objective To investigate the expression levels of microRNA-21(miR-21)in patients with pancreatic cancer(PC),and to analyze its correlation with clinical parameters,so as to evaluate its clinical significance in the disease.Methods A total of 53 patients with PC who were treated in our hospital from January 2017 to January 2019 were enrolled in the study.The expression levels of microRNA21 in cancer tissues and adjacent tissues were detected by quantitative real-time polymerase chain reaction(QR-PCR).Furthermore,the correlation between the expression levels and sprout RTK signaling antagonist-2(SPRY-2)as well as astrocyte elevated gene-1(AEG-1)was analyzed.Results The expression levels of miR-21 in cancer tissues(infiltrating,metastatic and atypical hyperplasia tissues)were significantly higher than those in adjacent tissues(P<0.05).The up-regulation of proliferation ability of panc-1 cells was positively correlated with the up-regulation of miR-21 expression(P<0.05).When the proliferation ability of MIAPaCa-2 cells was decreased,miR-21 expression levels were down-regulated(P<0.05).The inhibitory effects of DPI on invasion ability of PC were positively correlated with the drug concentration(P<0.05),so were the inhibitory effects of DPI on metastasis ability of PC tumor cells(P<0.05).DPI could obviously decrease the expression ability of RDS in primary tumor cells of PC(P<0.05),and MITO-TEMPO could block the expression of tetramethylpiperidine(P<0.05).Spearman rank correlation analysis showed that the expression levels of SPRY-2 protein were negatively correlated with those of miR-21(P<0.05),however,the expression levels of AEG-1 protein were not correlated with those of miR-21(P<0.05).Conclusion The expression levels of miR-21 in PC cancer tissues are obviously increased,which can be regarded as a new marker for PC diagnosis.
