摘　　要：目的建立UPLC方法,对医院制剂川芎定痛胶囊的相关成分进行定性定量分析,包括绿原酸、阿魏酸、延胡索乙素和欧前胡素。方法采用UPLC方法对四种主要成分对照品储备液进行测定,对医院制剂川芎定痛胶囊进行预处理,精密称定,50%甲醇提取液的UPLC分析采用HSS C18柱(2.1×150 mm,1.8μm),柱温35℃,以0.2%乙酸-乙腈为流动相梯度洗脱,在流速0.3 ml/min于波长254 nm检测。结果各成分具有良好线性关系(R2≥0.999),平均回收率在95.62%~102.67%(RSD在2.33%~3.19%)。结论建立UPLC川芎定痛胶囊主要有效成分含量测定的分析方法,提高原有川芎定痛胶囊的质量标准,进行多指标、多组分的含量测定,进一步正确、有效的评价川芎定痛胶囊质量,确保中药药效。Objective To establish an UPLC method to qualitatively and quantitatively analyze the relevant components of the Chuanxiongdingtong capsules prepared by hospital,including chlorogenic acid,ferulic acid,tetrahydropalmatine and imperatorin.Methods The UPLC method was used to determine the four main components of the reference substance stock solution.The Chuanxiongdingtong capsules prepared by hospital were pretreated and accurately weighed.The UPLC analysis of the 50%methanol extract was performed on a HSS C18 column(2.1×150mm,1.8μm),with column temperature 35℃,gradient elution with 0.2%acetic acid-acetonitrile as mobile phase,which were detected at a flow rate of 0.3ml/min at a wavelength of 254nm.Results The every component had a good linear relationship(R2≥0.999),and the average recovery rate was 95.62%~102.67%(RSD:2.33%~3.19%).Conclusion The UPLC method to detect main active ingredient contents of Chuanxiongdingtong capsules has beeen eastiblished,which can improve the original quality standard,carry out multi-indexes and multi-component content determination,and further correctively and effectively evaluate the quality of Chuanxiongdingtong capsules,so as to ensure the efficacy of Chinese medicine.
