摘 要:初步探讨山奈酚(Kaempferol,Kae)对肝癌细胞的抑制作用以及其影响肝癌细胞凋亡的相关机制。MTT法研究Kae对肝癌细胞活性的影响;镜下观察Kae处理后肝癌细胞形态和密度;通过TUNEL染色法研究Kae处理后肝癌细胞凋亡程度;通过免疫荧光实验和Western blot研究Kae对肝癌细胞自噬的影响。与对照组相比,Kae可显著抑制肝癌细胞活性,增加其肝癌细胞凋亡率,并且与Kae的浓度呈明显的相关性;Kae可显著降低肝癌细胞胞体的体积和细胞数目;TUNEL染色结果显示,与对照组相比Kae可明显抑制肝癌细胞活性,增加其细胞凋亡率;免疫荧光实验结果显示,Kae处理后肝癌细胞LC3表达水平较对照组相比显著升高;Western blot结果显示Kae处理后肝癌细胞的自噬相关蛋白p53、Bcl-2和LC3-Ⅱ/LC3-Ⅰ比值较对照组相比均明显升高。本研究发现Kae可通过可提高肝癌细胞自噬水平而影响其凋亡,同时作者的研究为肝癌的治疗提供了新的靶点和思路。To investigate the inhibitory effect of Kaempferol(Kae)on hepatocellular carcinoma cells and the related mechanism of its effect on apoptosis,MTT assay was used to study the effect of Kae on the cell activity of hepatoma cells.The morphology and density of HCC cells treated with Kae were observed under microscope.The apoptosis degree of HCC cells treated with Kae was studied by TUNEL staining.The effect of Kaeon autophagy of hepatoma cells was studied by immunofluorescence assay and Western blot.Compared with the control group,Kae could significantly inhibit the cell activity of HCC cells and increased the apoptosis rate of HCC cells,and it was significantly correlated with the concentration of Kae.Kae could significantly reduce the volume and number of hepatoma cells.TUNEL staining showed that Kae could significantly inhibit the cell activity and increased the apoptosis rate of HCC cells compared with the control group.The results of immunofluorescence assay showed that the expression level of LC3 in HCC cells after Kae treatment was significantly higher than that of the control group.Western blot results showed that the ratio of autophagyrelated proteins p53,Bcl-2 and LC3-Ⅱ/LC3-Ⅰ in HCC cells treated with Kae was significantly higher than that in the control group.In this study,it was found that Kae could affect the apoptosis of HCC cells by increasing the level of autophagy.Meanwhile,our study provided a new target and idea for the treatment of HCC.
