卵巢癌患者HPV16/18感染与抑癌基因Rap1GAP表达的关系

摘　　要：目的探讨卵巢癌患者人乳头瘤病毒16/18(HPV16/18)感染与抑癌基因Rap1GAP表达的相关性。方法回顾性选取2014年3月-2019年10月河南大学淮河医院妇科收治的200例卵巢癌患者作为研究对象,依据患者是否发生HPV16/18感染将其划分为感染组(n=106)和非感染组(n=94),检测和比较两组患者卵巢组织的Rap1GAP基因表达情况,分析卵巢癌患者HPV16/18感染和Rap1GAP基因表达之间的相关性。结果HPV16/18感染组Rap1GAP mRNA相对含量和表达率均低于非感染组(P<0.05),且Rap1GAP mRNA在卵巢癌中的表达与HPV16/18感染呈负相关性(r=-0.613,P=0.142)。200例卵巢癌患者中,HPV16/18感染与患者是否合并有糖尿病、肿瘤分化程度、性伴侣数、有无异体输血和Rap1GAP mRNA表达率有关(P<0.05);多因素分析结果显示,肿瘤低分化、性伴侣数≥2个和Rap1GAP mRNA表达(-)是卵巢癌患者HPV16/18感染的影响因素(P<0.05)。结论抑癌基因Rap1GAP在卵巢癌HPV16/18感染患者中的表达率和相对含量均较低,其与卵巢癌HPV16/18感染呈负相关性,Rap1GAP可能共同参与了卵巢癌的发生、发展过程。OBJECTIVE To explore the correlation between human papilloma virus 16/18(HPV16/18)infection and the expression of tumor suppressor gene Rap1GAP in patients with ovarian cancer.METHODS A retrospective study was performed on the 200patients with ovarian cancer admitted to Gynaecology Department of Huaihe Hospital of Henan University between Mar.2014and Oct.2019.According to presence or absence of HPV16/18infection,they were divided into the infection group(n=106)and non-infection group(n=94).The expression of Rap1GAP gene in ovarian tissues was detected and compared between the two groups.The correlation between HPV16/18infection and expression of Rap1GAP gene in patients with ovarian cancer was analyzed.RESULTS The test results showed that the relative expression and expression rate of Rap1GAP mRNA in the infection group were significantly lower than those in non-infection group(P<0.05).The expression of Rap1GAP mRNA in ovarian cancer was negatively correlated with HPV16/18infection(r=-0.613,P=0.142).Among the 200patients with ovarian cancer,the factors for HPV16/18infection was related to diabetes,tumor differentiation degree,number of sexual partners,allogeneic blood transfusion and expression rate of Rap1GAP mRNA(P<0.05).Multivariate Logistic regression analysis showed that low differentiation of tumors,sexual partners≥2and Rap1GAP mRNA(-)were important risk factors of HPV16/18infection in patients with ovarian cancer.CONCLUSION The expression rate and relative expression level content of tumor suppressor gene Rap1GAP are low in ovarian cancer patients with HPV16/18infection,which is negatively correlated with HPV16/18infection.Rap1GAP may participate in the occurrence and development of ovarian cancer.
