摘 要:目的研究黄精中酚类成分对二氧化硅纳米颗粒导致非小细胞肺癌细胞系(A549)细胞损伤的逆转和保护作用。方法利用硅胶柱色谱、ODS柱色谱、高效液相色谱等色谱手段对黄精中酚类成分进行分离纯化,经波谱数据分析鉴定其结构;建立体外二氧化硅颗粒诱导的A549损伤模型,采用MTT法测试酚类成分对受损细胞的保护作用。结果与结论从黄精根茎的体积分数为70%和90%的乙醇提取物中分离得到46个化合物,化合物3、4、6、8、13~15、17~21、23、24、26~28、30~33、35~37、46表现出显著的细胞保护作用,并能降低ROS的释放,提示其对细胞的保护作用机制可能与抗氧化有关。A549 cell is widely used to study the occurrence,development,diagnosis and treatment of lung cancer,and it is also used to study the occurrence,differentiation,structure and function of the normal alveolar epithelial cells typeⅡ.Chemoprevention is a promising strategy to control the damage of A549 cell.Natural products have been known as one of the most important resources of anticancer agents.Previous activity-screening research focused on the phenolic fraction(small polarity extract)of 70%and 90%ethanol extract of rhizomes of Polygonatum sibiricum.A continuous search for lung cancer chemopreventive agents resulted in the characterization of 46 compounds,which were separated and purified by different chromatographic methods(silica gel,ODS,HPLC,et al).We performed the chemopreventive activity of the extracts and isolates on SiO_(2)nanoparticles-induced A549 cell damages,and 25 components,including 3,4,6,8,13-15,17-21,23,24,26-28,30-33,35-37,and 46 presented significant chemopreventive effects with ED_(50);values from 1.4 to 14.0μmol·L^(-1),outperformed than positive control(resveratrol,14.7μmol·L^(-1)).ROS assays were further used to examine protective effects of isolates by the reduction of ROS production.
