摘  要:目的 探讨儿童免疫力低下与维生素A(vitamin A,VA)水平相关性,并分析与β-胡萝卜素-15,15.-单加氧酶(β,β-carotene-15,15.-monooxygenase,BCMO1)基因多态性的相关性,揭示BCMO1基因多态性与免疫力低下儿童的相关性。方法 根据T淋巴细胞亚群检测,将小孩分为免疫低下组和健康儿童组,每组各40例。采集各组外周血标本,收集临床资料。采用高效液相色谱法(HPLC)测定血清VA水平。设计引物用等位基因特异性PCR检测BCMO1多态性位点rs6564851、rs12934922和rs7501331基因型。结果 免疫低下组与健康儿童组比较,免疫低下组外周血清中VA水平较健康儿童组低,差异有统计学意义(P<0. 05)。免疫低下组感冒次数多于健康儿童组(P<0. 05),感冒次数与维生素A水平呈负相关。免疫低下组与健康儿童组比较,BCMO1多态性位点rs7501331、rs12934922基因型无明显差异,而rs6564851基因型在2组的分布差异有统计学意义(P=0. 04)。结论 免疫低下组儿童外周血中VA水平显著低于健康儿童组,BCMO1的3个SNPs中,rs6564851基因的多态性可能与儿童免疫相关疾病易感性有关,而rs7501331、rs12934922则与免疫相关疾病易感性无关。Objective To explore the relationship between low imunity of children and vitamin A(vitamin A,VA)levels andβ-carotene-15,15'-mooosygenase(β,β-carotene-15,15'-monooxygenase,BCMO1)gene polymorphism.The correla-tion of morphological expression reveals the correlation between VA metabolie enzyme polymorphism and children with weakened immunity.Methods According to the dection of T lymphocyte subsets,children were divided into immunocompromised group and healthy child group,with 40 cases in each group.Peripheral blood samples and elinical data were ollected.The ser-um VA level was determined by HPLC.Primers were designed to detect BCMO1 polymophie loci rs6564851,rs12934922 and ns7501331 genotypes by allele-specific PCR.Results Compared with the immunocompromised group and the healthy child group,the VA level in the peripheral serum of the immunocompromised group was lower than that of the healthy child group,and the diferences were statistically significanl(P<0.05).The number of colds in the immunocompromised group was higher than that in the healthy children group(P<0.05).The number of colds and vitamin A levels had a negative correlation.The rs7501331 and rs12934922 polymorphie loci of BCMO1 showed no significant difference between the immune-compromised group and the healthy group,while the distribution of RS6564851 showed significant difference between the two groups,and the difference was saistically signifieant(P=0.04).Conclusion'The level of vitamin A in the immunocompromised group is lower than that in the healthy child group.In the three SNPs of BCMOl,polymorpliism of rs6564851 gene may play a role in the pathogenesis of immune-related diseases,while the polyniorphism of rs7501331 and is 12934922 gene has nothing to do with children's susceptibility to colds.
