摘　　要：目的:探讨lncRNA MRCCAT1在乳腺癌组织中的表达,以及下调该基因对人乳腺癌MCF-7细胞增殖和侵袭的影响。方法:收集2018年6月至2019年12月在郑州大学第二附属医院行手术治疗的乳腺癌患者95例,培养MCF-7细胞并分为si-MRCCAT1组、si-NC组和空白组,分别转染MRCCAT1干扰序列、阴性对照序列和不作任何处理,实时荧光定量PCR术检测乳腺癌和癌旁组织以及细胞中MRCCAT1表达,MTT法检测细胞增殖活力,Transwell法检测细胞迁移和侵袭能力。结果:乳腺癌组织中MRCCAT1表达量为2.42±0.17,高于癌旁组织中的1.00±0.11,差异有统计学意义(P<0.001);乳腺癌组织中MRCCAT1表达量与PR、HER-2、组织学分级、TNM分期和淋巴结转移有关(P<0.05);si-MRCCAT1组细胞中MRCCAT1表达量低于si-NC组和空白组(P<0.05),si-MRCCAT1组24、48、72和96 h时细胞吸光度A值、迁移细胞数和侵袭细胞数均低于si-NC组和空白组(P<0.05)。结论:乳腺癌组织中MRCCAT1基因表达升高,且与恶性进展病理指标有关,干扰MCF-7细胞中MRCCAT1基因的表达可降低细胞增殖活性,抑制细胞迁移和侵袭能力。Objective:To investigate the expression of lncRNA MRCCAT1 in breast cancer tissues,and to explore the effect of down-regulation of this gene on the proliferation and invasiveness of human breast cancer MCF-7 cells.Methods:A total of 95 cases of patients with breast cancer who underwent surgical treatment in the Second Affiliated Hospital of Zhengzhou University were collected from June 2018 to December 2019.MCF-7 cells were cultured and divided into the si-MRCCAT1 group,si-NC group and blank group,and were transfected with MRCCAT1 interference sequence,negative control sequence and without any treatment,respectively.The expressions of MRCCAT1 in breast cancer and adjacent tissues and cells were detected by using real-time fluorescence quantitative PCR.Cell proliferation activity was detected by using MTT method and cell migration and invasion ability were detected by using Transwell method.Results:The expression level of MRCCAT1 in breast cancer tissues was 2.42±0.17,which was higher than 1.00±0.11 in adjacent tissues,the difference was statistically significant(P<0.001).The expression level of MRCCAT1 in breast cancer tissues was related to PR,HER-2,histological grade,TNM stage and lymph node metastasis(P<0.05).The expression level of MRCCAT1 in cells in the si-MRCCAT1 group was lower than that in the si-NC group and blank group(P<0.05).The cell absorbance A values at 24,48,72 and 96 h,the number of migrating cells and the number of invading cells in the si-MRCCAT1 group were all lower than those in the si-NC group and blank group(P<0.05).Conclusion:The expression of MRCCAT1 gene in breast cancer tissues is increased,and it is related to the pathological indicators of malignant progression.Interfering with the expression of MRCCAT1 gene in MCF-7 cells can reduce cell proliferation activity and inhibit cell migration and invasion abilities.
