摘 要:目的:探讨胶质瘤相关癌基因1(GLI1)在胃癌组织中的表达及临床意义。方法:收集2014年1月至2014年12月我院胃肠外科收治的经病理确诊行根治性手术的78例胃癌患者肿瘤组织及其邻近的正常胃组织标本。采用免疫组化染色方法检测胃癌及其邻近正常胃组织中GLI1表达情况,分析GLI1的表达与胃癌患者临床病理特征之间的相关性。结果:GLI1蛋白在胃癌组织中表达显著高于邻近正常胃组织(75.6%vs 26.9%,P<0.001),且GLI1的表达与肿瘤大小、肿瘤浸润深度、脉管侵犯、TNM分期、淋巴结转移有关,差异有统计学意义(P<0.05),而与性别、年龄、肿瘤部位、分化程度、神经侵犯无关(P>0.05)。生存分析利用Kaplan-Meier法,结果提示胃癌患者中GLI1高表达组总体生存时间显著低于低表达组,二者差异有统计学意义(P<0.0001)。结论:GLI1在胃癌组织中异常高表达,参与胃癌的发生发展及预后,未来可能成为胃癌基因治疗的新靶点。Objective:To investigate the expression and clinical significance of GLI1 in gastric cancers.Methods:The expression of GLI1 protein in 78 cases of gastric cancer and adjacent normal gastric tissues from January 2014 to December 2014 were detected by immunohistochemical staining,and the correlation between GLI1 expression and clinicopathological characteristics of gastric cancer patients was analyzed.Results:The expression of GLI1 protein in gastric cancer tissues was significantly higher than that in adjacent normal gastric tissues(75.6%vs 26.9%,P<0.001),and the expression level of GLI1 was related to tumor size,depth of tumor invasion,vascular invasion,TNM stage and lymph node metastasis(P<0.05),but not with gender,age,tumor location,histological differentiation and neural invasion(P>0.05).Kaplan-Meier analysis showed that the overall survival time of gastric cancer patients with high expression of GLI1 was significantly lower than that of patients with low expression of GLI1(P<0.0001).Conclusion:The abnormal high expression of GLI1 in gastric cancer tissue may participate in the occurrence,development and prognosis of gastric cancer,and may become a new target of gene therapy for gastric cancer in the future.
