灯盏细辛对人脉络膜黑色素瘤细胞凋亡的影响及作用机制

摘　　要：目的研究灯盏细辛对人脉络膜黑色素瘤(CM)细胞凋亡的影响及作用机制。方法将体外培养的OCM-1细胞分为对照组(NC组,正常培养液培养)、灯盏细辛低浓度组(LC组,浓度为1μmol/L)、灯盏细辛中浓度组(MC组,浓度为10μmol/L)及灯盏细辛高浓度组(HC组,浓度为100μmol/L),均培养48 h。CCK-8法测定细胞增殖率,Hoechst 33258核染色法观查细胞核凋亡,Annexin V/PI检测细胞凋亡,Western blot法检测OCM-1细胞内Bax及Bcl-2蛋白表达。结果(1)细胞增殖率:与NC组比较,各组均明显降低,差异有统计学意义(P<0.05)。与LC组比较,MC组降低不明显,差异无统计学意义(P>0.05);HC组明显降低,差异有统计学意义(t=5.639,P=0.000)。HC组比MC组明显降低,差异有统计学意义(t=4.214,P=0.000)。(2)细胞凋亡率:Hoechst 33258染色结果显示灯盏细辛干预后,高荧光类圆形细胞核数量增多,以HC组最为明显;与NC组比较,各组均明显升高,差异有统计学意义(P<0.05)。与LC组比较,MC组升高不明显,差异无统计学意义(P>0.05),HC组升高明显,差异有统计学意义(t=11.140,P=0.000)。HC组比MC组明显升高,差异有统计学意义(t=11.020,P=0.000)。(3)Bax蛋白表达:与NC组比较,各组均明显升高,差异有统计学意义(P<0.05)。与LC组比较,MC组降低,HC组升高,差异均有统计学意义(t_(MC)=17.48,t_(HC)=19.433,均P=0.000)。HC组比MC组升高,差异有统计学意义(t=3.680,P=0.006)。(4)Bcl-2蛋白表达:与NC组比较,各组均明显降低,差异有统计学意义(P<0.05)。与LC组比较,MC组和HC组均降低,差异均有统计学意义(t_(MC)=3.043,P=0.016,t_(HC)=9.767,P=0.000)。HC组比MC组降低,差异有统计学意义(t=7.267,P=0.000)。结论灯盏细辛通过改变线粒体凋亡途径中凋亡相关信号分子Bax及Bcl-2的表达,浓度依赖性抑制OCM-1细胞增殖并促进细胞凋亡。OBJECTIVE To study the effect and mechanism of scutellarin on the apoptosis of human choroidal melanoma(CM)cells.METHODS In vitro cultured OCM-1 cells were divided into control group(NC group,cultured in normal medium),low concentration of scutellarin(LC group,concentration of 1μmol/L),middle concentration of scutellarin group(MC group,concentration of 10μmol/L)and high concentration of scutellarin group(HC group,concentration of 100μmol/L),cells were cultured for 48h.Cells proliferation rate were determined by CCK-8 method.Nucleus apoptosis were observed by Hoechst 33258 nuclear staining.Cells poptosis rate were detected by Annexin V/PI.Protein expressions of Bax and Bcl-2 in OCM-1 cells were detected by Western blot method.RESULTS(1)Cells proliferation rate:compared with NC group,results of other groups was significantly lower,the difference had significant difference(P<0.05).Compared with LC group,result of MC group decreased slightly,the difference had no statistical significance(P<0.05);HC group significantly decreased,the difference was statistically significant(t=5.639,P=0.000).HC group decreased significantly than MC group,the difference was statistically significant(t=4.214,P=0.000).(2)Cells apoptosis rate:Hoechst 33258 staining results showed that the number of high fluorescence round nuclei increased after scutellarin intervention,especially in HC group.Compared with NC group,the apoptosis rate in each group was significantly higher,the difference was statistically significant(P<0.05).Compared with LC group,MC group increased inapparently,the difference had no statistical significance(P<0.05);HC group increased significantly,the difference was statistically significant(t=11.140,P=0.000).HC group increased significantly than MC group,the difference was statistically significant(t=11.020,P=0.000).(3)Bax protein expression:compared with NC group,results of other groups increased significantly,the difference was statistically significant(P<0.05).Compared with LC group,result of MC group decreased,HC gro
