摘 要:目的评估基于VP4/VP2结合区基因序列对手足口病(hand,foot and mouth disease,HFMD)病原分型方法的准确性,建立快速简易的手足口病病原分型方法。方法用逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)对2020年云南省曲靖市送检的49株已用VP1区基因序列分型的肠道病毒进行VP4/VP2结合区基因扩增并测序,获得VP4/VP2结合区分型结果。将VP4/VP2结合区基因分型结果与VP1区分型结果进行一致性评价,利用Mega 5.2软件对毒株基因组序列进行分析。结果49株肠道病毒经VP4/VP2结合区基因序列分型,共分出5个型别,包括肠道病毒(enterovirus,EV)-A组48株4个型别,EV-B组1株1个型别,未分离到EV-C、EV-D。与VP1区分型结果比较,EV-A、EV-B组病毒分型符合率100%,但EV-B组病毒只检测到1株。结论基于VP4/VP2结合区的手足口病病原分型方法对EV-A组病毒能准确分型,对EV-B、EV-C和EV-D组病毒的分型准确率有待积累更多病毒型别进一步研究。Objective To assess the accuracy of the typing method based on VP4/VP2 junction region gene sequences and to establish a rapid and simple method for typing enteroviruses from hand,foot and mouth disease(HFMD).Methods VP4/VP2 junction gene of 49 enteroviruses typed by VP1 gene sequences were amplified by RT-PCR and sequenced to obtain typing results by VP4/VP2 gene.Consistency between typing results by VP4/VP2 and VP1 genes was assessed to determine the accuracy of typing by VP4/VP2.The Mega 5.2 software was used to compare the sequences and construct a genetic evolutionary tree.Results All of the 49 total strains typed by VP1 gene,5 serotypes including 48 strains of species EV-A(4 serotypes)and 1 strain of species EV-B(1 serotype)were typed by VP4/VP2 region.Species EV-C and EV-D viruses were not typed.The consistency rate of 100%between VP4/VP2 and VP1 gene region for species A and B was obtained.However,only 1 strain of species B virus was obtained.Conclusions VP4/VP2 typing is accurate for enterovirus species A viruses,the typing accuracy of species B,C and D viruses awaits for further verification until more viruses are obtained.
