摘 要:肿瘤坏死因子-α(Tumor necrosis factorα,TNF-α)是一种具有多种生物学活性的细胞因子。为研究高糖高脂饲料对草鱼不同组织中TNF-α及TRAF1/TRAF2(TNF receptor-associated factor 1/TNF receptor-associated factor 2)表达的影响,本研究主要采用qRT-PCR的方法检测了高糖高脂饲料饲喂草鱼65 d后,草鱼脂肪、肌肉和肝脏组织中TNF-α、TRAF1和TRAF2基因转录水平的变化。结果显示,高糖高脂饲料可诱导草鱼肝脏脂肪大量蓄积。与对照组相比,饲喂高糖高脂饲料组的草鱼脂肪组织中TNF-α,TRAF1和TRAF2的表达量均显著升高(P<0.05),但三者在肌肉中的表达量都显著降低(P<0.05),在肝脏中仅TRAF1的表达量出现显著升高(P<0.05),TNF-α和TRAF2的表达量均没有显著性变化(P>0.05)。结果表明,在高糖高脂饲料应激下,TNF-α,TRAF1和TRAF2在三种组织中存在明显不同的表达模式,同一个基因在不同组织中的表达变化也不同。这为进一步研究TNF-α及其TNF受体相关因子在调节草鱼脂质代谢和免疫等方面的功能提供了理论依据。Tumor necrosis factor-α(TNF-α)is a cytokine with multiple biological activities.In order to explore the effect of high carbohydrate and high fat diet on the gene expression of TNF-α,TRAF1 and TRAF2 in different tissues of grass carp,the transcription levels changes of TNF-α,TRAF1 and TRAF2 in adipose,liver and muscle tissue of grass carp were detected by qRT-PCR method after 65 d feeding with high carbohydrate and high fat diet.The results showed that high carbohydrate and high fat diet significantly caused liver fat accumulation in large quantities.Compared with expression index of the control group,after high carbohydrate and high fat feeding,the expressions index of TNF-α,TRAF1 and TRAF2 in adipose tissue were significantly increased(P<0.05),and the expression index of these related genes in muscle tissue were significantly reduced(P<0.05).In liver,compared with the control group,only the expression level of TRAF1 was significantly increased in the high carbohydrate and high fat group(P<0.05),while the expression levels of TNF-αand TRAF2 were not significantly changed.Overall,the results indicated that the expressions of TNF-α,TRAF1 and TRAF2 were obviously different in liver,muscle and adipose tissue under the stress of high carbohydrate and high fat diet,and the expression changes of the same gene in different tissues were also different.Moreover,this study can provide a basis for further research on the mechanism of TNF-αand its TNF receptor related factors in regulating lipid metabolism and immunity in grass carp.
