摘  要:目的研究羟基红花黄色素A(HSYA)对离体大鼠大脑中动脉(MCA)的肌原性作用及其作用机制。方法从大鼠脑组织中分离得到MCA血管环,并分离其血管平滑肌细胞。将同一根血管的不同节段随机分为5组:对照组(生理盐水)、HSYA-1组、HSYA-2组、HSYA-3组和HSYA-4组(HSYA浓度分别对应3,10,30,100μmol·L^(-1));将同一根血管的不同节段随机分为2组:内皮完整组和去内皮组;用同一根血管自身前后对照,分为2组:HSYA组和G?6983+HSYA组,上述实验均用微血管张力法检测大鼠MCA血管环的舒张功能。将同一只大鼠MCA平滑肌细胞分为4组:对照组(生理盐水)、HSYA-1组、HSYA-2组和HSYA组-3(HSYA浓度分别对应10,30,100μmol·L^(-1));将同一只大鼠MCA平滑肌细胞分为4组:对照组(生理盐水)、HSYA组、G?6983组和G?6983+HSYA组,上述实验均用全细胞膜片钳记录法检测MCA平滑肌细胞的Kv电流。结果对照组、HSYA-1组、HSYA-2组、HSYA-3组和HSYA-4组的舒张KCL预收缩的MCA百分比分别为(100.00±0.00)%,(98.32±1.39)%,(92.07±3.92)%,(59.40±5.07)%和(35.89±5.62)%,各组舒张U46619预收缩的MCA百分比分别为(100.00±0.00)%,(98.82±0.70)%,(92.57±5.06)%,(59.23±7.21)%和(40.22±6.67)%,HSYA-2组、HSYA-3组、HSYA-4组与对照组比较,差异均有统计学意义(P<0.05,P<0.01);内皮完整组与去内皮组比较,各剂量组HSYA舒张预收缩大鼠MCA血管环的作用差异无统计学意义(均P>0.05);100μmol·L^(-1)HSYA组和G?6983+100μmol·L^(-1)HSYA组舒张KCL预收缩的MCA百分比分别为(34.93±4.91)%和(58.39±6.57)%,这2组舒张U46619预收缩的MCA百分比分别为(37.95±4.13)%和(53.59±6.00)%,上述指标,两组间比较差异均有统计学意义(均P<0.01);对照组、HSYA-1组、HSYA-2组和HSYA-3组MCA平滑肌细胞Kv电流最大电流密度分别为(41.24±4.34),(49.48±5.56),(67.94±5.61)和(78.84±3.79)pA/pF,各浓度HSYA组与对照组比较,差异均有统计学意义(P<0.05,P<0.01);对照组、HSYA组、G?6983组和G?6Objective To investigate the myogenic effect of Hydroxysafflor yellow A on the isolated rat middle cerebral artery(MCA)rings and its underlying mechanisms.Methods MCA rings and its vascular smooth muscle cells were obtained from rat MCA.Different segments of the same MCA were randomly divided into five groups:control group(normal saline),HSYA-1 group,HSYA-2 group,HSYA-3 group and HSYA-4 group(3,10,30,100μmol·L^(-1)HSYA);different segments of the same vessel were randomly divided into two groups:endothelium-intact group and endothelium-denuded group;the same blood vessel was used before and after control and was divided into two groups:HSYA group and G?6983+HSYA group,the vascular myogenic tone of rat MCAs experiments was recorded with a wire myograph.The smooth muscle cells of the same rat were divided into four groups:control group(normal saline),HSYA-1 group,HSYA-2 group and HSYA-3 group(10,30,100μmol·L^(-1)HSYA);the smooth muscle cells of the same rat were divided into 4 groups:control group(normal saline),HSYA group,G?6983 group,G?6983+HSYA group.Kv currents of freshly isolated rat MCA smooth muscle cells experiments were detected by whole-cell patch clamp technique.Results The diastolic percentage of control group,HSYA-1 group、HSYA-2 group、HSYA-3 group and HSYA-4 group in KCl precontracted MCA rings were(100.00±0.00)%,(98.32±1.39)%,(92.07±3.92)%,(59.40±5.07)%and(35.89±5.62)%;in U46619 precontracted MCA rings were(100.00±0.00)%,(98.82±0.70)%,(92.57±5.06)%,(59.23±7.21)%and(40.22±6.67)%,there were significant differences between HSYA-2,-3,-4 groups and control group(P<0.05,P<0.01).The relaxation effect of HSYA(3-100μmol·L^(-1))on precontracted MCA showed no statistical difference between endothelium-intact and endothelium-denuded group(P>0.05).The diastolic percentage of 100μmol·L^(-1)HSYA group and G?6983+100μmol·L^(-1)HSYA group in KCl precontracted MCA rings were(34.93±4.91)%and(58.39±6.57)%;in U46619 precontracted MCA rings were(37.95±4.13)%and(53.59±6.00)%.There were significant
