结直肠癌组织中LncRNA TUSC7和p53的表达及临床意义

摘　　要：目的:检测结直肠癌组织中长链非编码RNA抑癌候选基因7(LncRNA TUSC7)和p53的表达水平,并探讨其对结直肠癌的临床意义。方法:选取2016年2月—2017年2月收治的76例结直肠癌患者术后留取组织标本作为观察组,并选择观察组中距肿物超过5 cm的切缘正常结肠黏膜组织76份作为对照组。以终点事件发生作为预后不良的观察指标,将观察组患者分为预后不良组25例和预后良好组49例(2例失访)。采用实时荧光定量PCR(qRT-PCR)法检测组织中LncRNA TUSC7、p53表达水平;分析LncRNA TUSC7、p53水平与结直肠癌临床病理特征的关系;Pearson分析LncRNA TUSC7、p53表达水平的相关性;多因素Cox回归分析影响结直肠癌预后的因素。结果:观察组LncRNA TUSC7、p53表达水平低于对照组(P<0.05);LncRNA TUSC7、p53表达与肿瘤直径、AJCC分期、组织分化程度、淋巴结转移有关(P<0.05);LncRNA TUSC7、p53表达水平呈正相关(r=0.585,P<0.001);预后不良组LncRNA TUSC7、p53水平低于预后良好组(P<0.05);LncRNA TUSC7、p53水平是影响结直肠癌预后的独立危险因素(P<0.05)。结论:结直肠癌组织中LncRNA TUSC7、p53水平呈低表达,与临床分期等临床病理特征密切相关,且是影响结直肠癌预后的独立危险因素。Objective:To detect the expression levels of long non-coding RNA tumor suppressor candidate 7(LncRNA TUSC7)and p53 in colorectal cancer tissues,and to explore their clinical significance.Methods:From February 2016 to February 2017,tissue samples from 76 patients with colorectal cancer were selected as the observation group after operation,and 76 normal colonic mucosa tissues with a distance of more than 5 cm from the tumor of patients in the observation group were selected as the control group.The expression levels of LncRNA TUSC7 and p53 in tissues were detected by real-time fluorescent quantitative PCR(qRT-PCR);the relationships between the levels of LncRNA TUSC7 and p53 and the clinicopathological features of colorectal cancer were analyzed;Pearson was used to analyze the correlation between the expressions of LncRNA TUSC7 and p53;Multivariate Cox regression was used to analyze the prognostic factors of colorectal cancer.Results:The levels of LncRNA TUSC7 and p53 in the observation group were lower than those in the control group(P<0.05);the expressions of LncRNA TUSC7 and p53 were related to tumor diameter,AJCC stage,degree of tissue differentiation and lymph node metastasis(P<0.05);there was a positive correlation between the expression levels of LncRNA TUSC7 and p53(r=0.585,P<0.001);the levels of LncRNA TUSC7 and p53 in the poor prognosis group were lower than those in the good prognosis group(P<0.05);the levels of LncRNA TUSC7 and p53 were independent risk factors for the prognosis of colorectal cancer(P<0.05).Conclusions:The expressions of LncRNA TUSC7 and p53 in colorectal cancer tissues are low,which are closely related to clinical stage and other clinicopathological features,and are independent risk factors for the prognosis of colorectal cancer.
