摘 要:目的利用癌症基因组图谱(the cancer genome atlas, TCGA)数据库分析胃腺癌组织中赖氨酸羟化酶3(procollagen-lysine, 2-oxoglutarate 5-dioxygenase,PLOD3)基因表达及其潜在临床价值。方法使用UALCAN数据库(http://ualcan.path.uab.edu/)分析TCGA中PLOD3基因在胃腺癌及癌旁组织间表达差异;利用Kaplan-Meier模型分析PLOD3基因表达与胃腺癌患者预后相关性;利用LinkedOmics数据库筛查PLOD3相关基因;应用免疫组织化学验证胃腺癌与癌旁正常胃黏膜组织PLOD3来源与分布差异。结果数据库资料表明,胃腺癌组织PLOD3基因表达高于正常胃组织(P<0.001);伴有p53基因突变的胃腺癌患者PLOD3基因表达高于无p53基因突变患者(P<0.001);PLOD3基因表达与胃腺癌患者生存期相关(HR=1.360,95%CI:1.150~1.610,P<0.001);PLOD3基因表达与AP1S1、LRWD1和SLC12A9等基因表达呈正相关(均有P<0.001),与CGGBP1、ZNF561和MIER1等基因表达呈负相关(均有P<0.001);免疫组化染色结果进一步证实胃腺癌组织PLOD3蛋白表达高于癌旁正常胃黏膜组织(P=0.031)。结论 PLOD3可能成为评价胃腺癌预后以及胃腺癌靶向治疗的有效新靶点。Objective To analyze the expression of procollagen-lysine,2-oxoglutarate 5-dioxygenase(PLOD3)and its clinical significance in gastric adenocarcinoma by means of the cancer genome atlas(TCGA)database.Methods The UALCAN database(http://ualcan.path.uab.edu/)was used to analyze differentiated expression of PLOD3 gene in gastric adenocarcinoma tissues and normal gastric tissues in TCGA database.Kaplan-Meier model was designed to analyze the correlation between the prognosis of patients with gastric adenocarcinoma and the expression of PLOD3.LinkedOmics database was implemented to analyze the genes related to PLOD3.Immunohistochemistry was carried out to verify the expression difference of PLOD3 in gastric adenocarcinoma and adjacent normal gastric tissues.Results The database analysis revealed that the expression level of PLOD3 gene in gastric cancer tissues was higher than that in normal gastric tissues(P<0.001).Expression of PLOD3 gene in gastric adenocarcinoma with p53 mutation was higher than the cases without p53 mutation(P<0.001).PLOD3 gene expression was significantly correlated with the survival of patients with gastric adenocarcinoma(HR=1.360,95%CI:1.150-1.610,P<0.001).PLOD3 expression was positively correlated with the genes,such as AP1S1,LRWD1,and SLC12A9(all P<0.001),and negatively correlated with the genes,such as CGGBP1,ZNF561 and MIER1(all P<0.001).The immunohistochemical observation further demonstrated that more PLOD3 protein was expressed in gastric adenocarcinoma tissues than the adjacent noncancerous gastric tissues(P=0.031).Conclusion PLOD3 would become a valuable biomarker for evaluating the prognosis and targeted therapy for patients with gastric adenocarcinoma.
