摘 要:目的:探讨口腔癌组织中肿瘤坏死因子受体相关因子6(TNF receptor-associated factor 6,TRAF6)的表达及其对细胞自噬、凋亡及NF-κB/c-jun信号途径的影响。方法:qRT-PCR和Western blot检测TRAF6在口腔癌组织和口腔癌细胞系中的表达;qRT-PCR和Western blot检测沉默TRAF6在SCC-25细胞中的表达;EdU实验检测SCC-25细胞的增殖能力;Western blot检测SCC-25细胞中LC3Ⅱ/LC3 I比例;细胞免疫荧光染色检测SCC-25细胞中自噬体数目;流式细胞术检测SCC-25细胞的凋亡率;Western blot检测SCC-25细胞中NF-κB和c-jun蛋白的表达。结果:TRAF6在口腔癌组织和口腔癌细胞系中高表达(P<0.001);沉默TRAF6使口腔癌细胞系中TRAF6的表达降低;沉默TRAF6使SCC-25细胞的增殖能力下降(P<0.01)、LC3Ⅱ/LC3 I比例下降(P<0.001)和自噬体数目减少(P<0.001);沉默TRAF6使SCC-25细胞的凋亡率升高(P<0.01),NF-κB和c-jun蛋白磷酸化程度降低(均P<0.001)。结论:TRAF6在口腔癌组织和口腔癌细胞中高表达,沉默TRAF6抑制SCC-25细胞自噬,增强SCC-25细胞的凋亡率及抑制NF-κB/c-jun信号途径的活性。Objective:To investigate the expression of TRAF6 in oral cancer tissues and its effect on autophagy,apoptosis and NF-κB/c-jun signaling pathway in oral cancer cells.Methods:qRT-PCR and Western blot were used to detect the expression of TRAF6 in oral cancer tissues and oral cancer cell lines.qRT-PCR and Western blot were used to detect the expression of silent TRAF6 in SCC-25 cells.EdU test was used to detect the proliferation ability of SCC-25 cells.Western blot was used to detect the ratio of LC3Ⅱ/LC3 I in SCC-25 cells.Cellular immunofluorescence staining was used to detect the number of autophagosomes in SCC-25 cells.Cell flow cytometry was used to detect the apoptosis rate of SCC-25 cells.Western blot was used to detect the expression of NF-κB and c-jun protein in SCC-25 cells.Results:TRAF6 was highly expressed in oral cancer tissues and oral cancer cell lines(P<0.001).Silencing TRAF6 reduced the expression of TRAF6 in oral cancer cell lines.Silencing TRAF6 decreased the proliferation ability of SCC-25 cells(P<0.01),the ratio of LC3Ⅱ/LC3 I(P<0.001),and the number of autophagosomes(P<0.001).Silencing TRAF6 increased the apoptotic rate of SCC-25 cells(P<0.01),and decreased the phosphorylation of NF-κB and c-jun(both P<0.001).Conclusion:TRAF6 was highly expressed in oral cancer tissues and oral cancer cells.Silencing TRAF6 inhibited autophagy of SCC-25 cells,enhanced the apoptosis rate of SCC-25 cells and inhibited the activity of NF-κB/c-jun.
