摘  要:目的探讨骨髓间质干细胞(BMSCs)移植联合超声介入治疗犬急性脑梗死模型的价值。方法18只杂种犬,犬龄6~10年,体重5~20 kg,所有犬均建立急性脑梗死模型,随机数字表法平分为3组-模型组、介入组、干细胞组,介入组给予超声介入治疗,干细胞组给予BMSCs移植联合超声介入治疗,模型组不进行任何治疗处理。观察比较3组治疗第2周与第4周的mNSS神经功能评分、凝血酶原时间(PT)与纤维蛋白原(FIB);评价3组治疗第4周的脑血管造影总有效率;测定3组治疗第4周的海马组织Caspase-3、Bax蛋白相对表达水平。结果介入组、干细胞组治疗第2周与第4周的mNSS神经功能评分均低于模型组,且干细胞组低于介入组,差异均有统计学意义(P<0.05)。3组治疗不同时间点的PT与FIB等凝血纤溶功能指标在组内与组间对比差异均无统计学意义(P>0.05)。模型组、介入组、干细胞组治疗第4周的脑血管造影总有效率分别为100.0%、50.0%和16.7%,干细胞组总有效率显著低于模型组、介入组,差异有统计学意义(P<0.05)。介入组、干细胞组治疗第4周的海马组织Caspase-3、Bax蛋白相对表达水平低于模型组,且干细胞组低于介入组,差异均有统计学意义(P<0.05)。结论BMSCs移植联合超声介入治疗犬急性脑梗死能抑制海马组织细胞凋亡,且不会影响犬的凝血及纤溶状况,从而可提高总体治疗效果,改善神经功能状况。Objective To discuss the values of bone marrow mesenchymal stem cells(BMSCs)transplantation combined with ultrasound intervention in the treatment of acute cerebral infarction in dogs.Methods The 18 mongrel dogs,6-10 years old,5-20 kg weight,all dogs were established with acute cerebral infarction models and divided into 3 groups-model group,interventional group,and stem cell group according to the random number table method.The interventional group were given ultrasound interventional therapy,and the stem cell group were given BMSCs transplantation combined with ultrasound interventional therapy.The model group were not conducted any treatment treatment.The mNSS neural function score,prothrombin time(PT)and fibrinogen(FIB)of the 3 groups at the 2nd and 4th weeks of treatment were observed and compared;the total effective rate of cerebral angiography in the 3 groups at the 4th week of treatment was evaluated;The relative expression levels of Caspase-3 and Bax protein in hippocampus tissues of the 3 groups were determined at the 4th week of treatment.Results The mNSS neurological scores of the interventional group and the stem cell group were lower than the model group at the 2nd and 4th weeks of treatment,and the stem cell group were lower than those of the intervention group,the differences were statistically significant(P<0.05).The PT and FIB and other blood coagulation and fibrinolytic function indexes at different time points of the three groups of treatment compared were not statistically different within and among the groups(P>0.05).The total effective rates of cerebral angiography in the model group,interventional group,and stem cell group were 100.0%,50.0%,and 16.7%at the 4th week of treatment,respectively,the total effective rate of the stem cell group was significantly lower than that of the model group and intervention group,the differences were statistically significant(P<0.05).The relative expression levels of Caspase-3 and Bax protein in the hippocampus tissue of the interventional group and the ste
