摘  要:为了给今后应用于耐药性肺炎克雷伯菌感染的噬菌体生物制剂的开发提供依据,本试验分离鉴定多重耐药肺炎克雷伯菌裂解性噬菌体并进行遗传信息分析。以多重耐药肺炎克雷伯菌为宿主菌,从医院污水中分离肺炎克雷伯菌噬菌体并进行纯化、电镜观察形态特征、生物学特性分析、提取噬菌体DNA,进行全基因组测序,分析全基因组的结构特征,比较基因组分析其进化关系。结果显示:分离得到肺炎克雷伯菌裂解性噬菌体Kp-8329,属于有尾噬菌体目、长尾病毒科;基因组全长46907 bp,有116个开放阅读框,未检出tRNA和rRNA相关基因;噬菌体Kp-8329与大肠埃希菌属噬菌体有较为接近的进化关系。结果表明,Kp-8329是一种潜伏期短、裂解效率高、耐高温以及耐酸碱的噬菌体,可考虑用于耐药性肺炎克雷伯菌感染的噬菌体生物制剂的开发。To facilitate the development of phage biologics against drug-resistant Klebsiella pneumoniae infection,multi-drug-resistant K.pneumoniae lytic phage was isolated,identified and its genetic information was analyzed.Using K.pneumoniae as a host,K.pneumoniae lytic phage was isolated from hospital sewage,its morphological features were examined by electron microscopy,biological characteristics were determined,phage DNA was extracted,whole genome was sequenced,structural features of the whole genome were analyzed,and evolutionary relationship was established through genome comparison.The lytic phage Kp-8329 of K.pneumoniae was isolated,and shown to belong to the order Tailed phages and family Veroviridae.The genome is 46907 bp in length,with 116 open reading frames,but without tRNA and rRNA related genes.The bacteriophage Kp-8329 shares a relatively close evolutionary relationship with Escherichia coli phages.Kp-8329 is a phage that has short incubation period,high lysis efficiency,and resistance to high temperature,acid and alkali.It can be considered for the development of phage biologics against drug-resistant K.pneumoniae infection.
