摘 要:目的:探讨缺血性脑卒中气虚血瘀证与气滞血瘀证大鼠肠道菌群的差异。方法:44只SPF级7~9周龄SD大鼠随机分为4组。分别采用改良Z-longa线栓法制作大脑中动脉阻塞型缺血性脑卒中大鼠模型(MCAO组),力竭游泳结合线栓法制作缺血性脑卒中气虚血瘀证大鼠模型(QXY组),肾上腺素皮下注射联合多种混合刺激及线栓法制作缺血性脑卒中气滞血瘀证大鼠模型(QZY组),每组12只。另设假手术组(SOG组)8只。大鼠术后72h盲肠内容物进行16S rRNA基因测序检测肠道菌群,并进行生物信息学分析。结果:与SOG组比较,QXY组物种丰富度明显下降,厚壁菌门丰度显著降低,而变形杆菌门的丰度水平显著升高;QXY组变形杆菌门和疣微菌门丰度轻度升高,厚壁菌门轻度下降,但差异无统计学意义。与MCAO组比较,QXY组大鼠厚壁菌门及脱铁杆菌门丰度明显下降;QZY组脱铁杆菌门丰度明显下降。与QZY组比较,QXY组埃希-志贺菌属、考拉杆菌属、AListipes、粪杆菌属、Odoribacter、Sutterella等10个菌属丰度显著升高,而Parasutterella、Lachnospiraceae_UCG-001菌属丰度显著下降。LEFSE分析发现Sutterella菌属是QXY组大鼠肠道菌群特异性的主要菌属。结论:气虚血瘀证缺血性脑卒中大鼠肠道菌群与其他证型缺血性脑卒中大鼠存在显著性差异,提示肠道菌群可能参与缺血性脑卒中气虚血瘀证中医证候的形成。Objective:To investigate the difference of intestinal flora between qi deficiency and blood stasis syndrome and qi stagnation and blood stasis syndrome in rats with ischemic stroke.Methods:Forty-four SPF grade 7 to 9-week old SD rats were ran domly divided into 4 groups.The middle cerebral artery occlusion ischemic stroke rat model(MCAO group)was established by thread occlusion method;the rat model of qi deficiency and blood stasis syndrome in ischemia(QXY group)was established by exhaustive swimming combined with thread embolism and the rat model of qi stagnation and blood stasis syndrome(QZY group)was established by subcutaneous injection of epinephrine combined with various mixed stimulation and thread occlusion,respectively,12 rats in each group.The sham operation group(SOG group)was set up with 8 rats of all groups.The cecal contents were collected 72 h after operation.16 S rRNA gene sequencing was used to detect the intestinal flora,and bioinformatics analysis was performed.Results:Compared with SOG group,the species richness of QXY group decreased significantly,the Firmicutes abundance decreased significantly,and the Proteus abundance increased significantly;and the abundance of Proteus and verruca increased slightly,while Firmicutes decreased slightly,but there was no significant difference.Compared with MCAO group,the abundance of Firmicutes and deferrobacteria decreased significantly in QXY group;while in QZY group,the abundance of deferrobacterium decreased significantly.Compared with QZY group,the abundances of Shigella,koala,alistipes,faecalis,odoribacter and sutterella in QXY group were significantly increased,while the abundances of Parasutterella and Lachnospiraceae_UCG-001 were significantly decreased.LEFSE analysis showed that Sutterella was the main genus of intestinal flora in QXY group.Conclusion:There are significant differences between the intestinal flora of ischemic stroke rats with qi deficiency and blood stasis syndrome and other syndrome types of ischemic stroke rats,suggesting that th
